To develop the traditional national resource of our country, the thesis deals with the chemical consistituents, the optimum technics condition and the activities of extracting total flavonoids of Saxifraga tangutica Engl. which were used as Tibetan medicines in China.Twelve pure compounds were isolated by using chromatographic column on silica gel and polyamide polymer . The structures of eight compounds were elucidated on the basis of spectral evidence including NMR, IR, MS, UV, ID and chemical methods. Eight compounds were isolated from the hers plants for the first time and identified as: quercetin (1), quercetin-3-O-β-D-glucopyranoside (2), quercetin-3-O-β-D-glucopra -noside (3), quercetin-3-O-β-D-galactoside-7-O-β-D-glucoside (4),β-sitosterol (5), dausterol (6), nonacosane(7), hentriacontane (8).Flavonoids were rich in Saxifraga and they were thought as the activity compounds. To the further utilization of Saxifraga tangutica Engl., the studies were to confirm the optimum technics condition of extracting total flavonoids from Saxifraga tangutica Engl. assisted by ultrasound wave. Based on single factor test, the orthogonal experiments were conducted with 4 factors of ethanol concentration. The optimum technics conditions were made of 80% ethanol as extraction solvent, 1:25(g:mL) as material to ethanol ratio, 30min as ultrasonic extraction time, treating with ultrasonic for three times repeatedly at the temperature of 50oC and ultrasonic power of 100W . The total flavonoids extraction efficiency could be reached as high as 81.73mg/g .Thirdly, the studies were to confirm the pharmacologic effects, both anti-bacterial activity and anti-aging with the total flavonoids extracted efficiently. Using agar diffusion method and broth coubling dilution the anti-bacterial experements were shown that the flavonoids extraction were shownsignificant anti-bacterioustaic activity and weak anti-eumycete activity; the smallest kill-bacterial concentration were respectly 0.156, 0.313, 0.625 mg/mL to Staphylococcus aureus, Bacillus subtilis and Escherichia col-I; Then the total flavonoids were given to mice injected D-gal to study the ability of anti-aging when the contents of heyocuprein (SOD), glutathion peroxidase (GSH-Px) and malonaldehyde (MDA) were determined by D-galactoside. The ability of SOD and GSH-Px could be increased in the tissues and that of MDA could be decreased (P<0.01). So as to avoid the damage of the tissues caused by the freeradical over-oxidization; it may be an effective protective drug for the anti-aging.
