Study On Lipid Peroxidation And Anti-Oxidant Capacity In The Liver, Brain And Testis Tissues Of The Mice Exposured To Acrylonitrile

Objective To explore the effect of acrylonitrile (AN) on lipid peroxidation and activities of anti-oxidizes in the liver, brain and testicular tissues of the mice, in order to provide some theoretical data for protecting the health of occupational AN exposure population.Methods Two hundred adult male mice(6~8 weeks old) in SPF degree were randomly divided into 4 groups according to the body weight including the negative control group(normal saline),the low-dose AN group(1.25mg/kg),the medium-dose AN group(2.5mg/kg),the high-dose AN group(5mg/kg),each group has 50 mice. All mice were treated by daily intraperitoneal injection for 5 days, the injected dose of all dose groups was 10ml/kg. All mice were sacrificed by cervical dislocation in batch at the 7th,14th,21th,28th,35th day after the first administration.(i)Liver, brain and testis were taken to be weighted and calculated the organic indexes. (ii)The content of ALT, AST, ALT/AST, ALP, TP, ALB, GLO, A/G and ChE in rerum were measured. (iii)The activities of glutathione peroxidase(GSH-Px),superoxide dismutase(SOD),catalase(CAT),cholinesterase(ChE) and the levels of glutathione (GSH),malondialdehyde(MDA) in the brain tissue, the activities of GSH-Px, SOD, Na+-K+-ATPase, Ca2+-ATPase, Mg2+-ATPase and the levels of MDA in the liver tissues, the activities of T-AOC, SOD and the levels of GSH, MDA in testicular homogenate of the experimented mice were determined respectively with spectrophotometric assay.Results (i)There were no statistical significances in all kinds of organic indexes among all groups(P>0.05). (ii) Blood biochemical criterion:Compared with the negative control group, AST,ALT/AST,ALP,ALB,GLO in low concentration AN groups,AST in medium concentration AN groups and AST,ALP,ALB,GLO,ChE in high concentration AN groups were significantly higher than that negative control group in 7-day group. (iii)The lipid peroxidation and activities of anti-oxidizes Parameters in the brain tissue:Compared with the negative control group, the levels of GSH were statistically decreased in the medium and high concentration AN groups of 28-day group.The levels of MDA in the medium and high concentration AN groups of 7-day group and in the low,medium,high concentration AN groups of 21-day group were significantly higher than the negative control group.The activities of GSH-Px were statistically increased in the low and high concentration AN groups of 21-day group,and the activities of CAT were increased in the low,medium and high concentration AN groups of 21-day group compared with negative control group. (iv)The lipid peroxidation and activities of anti-oxidizes Parameters in the liver tissue: Compared with the negative control group,the levels of MDA were statistically increased in the medium,high concentration AN groups of 28-day group and in the high concentration AN groups of 35-day group.The activities of SOD, GSH-Px were significantly higher than the negative control group in the high concentration AN group of 28-day group. At different ending times ATPase activeties had no statistic significance among every dose group. (v)The lipid peroxidation and activities of anti-oxidizes Parameters in the testis tissue:Compared with the negative control group,the levels of MDA were statistically increased in the medium concentration AN group of 21-day group and in the medium,high concentration AN groups of 35-day group. The levels of GSH were increased in the medium,high concentration AN groups of 7-day group and in the high concentration AN group of 28-day group compared with the negative control group. The activity of T-AOC in high concentration AN group of 21-day group was significantly higher than the negative control group. (vi)The levels of MDA were incresed obviously in 21-day group of the brain tissue and in 35-day group of the liver tissue.The level of MDA in the medium concentration AN group was incresed obviously in the testis tissue.Conclusion (i)Acrylonitrile exposure could impair the balance of antioxidant system and induce oxidative damage in the brain,liver,testis tissues. Effect of Lipid peroxidation in the brain tissue was earlier and more serious than that in the liver tissues, testis was sensitive to AN in the medium concentration AN group (2.5mg/kg).In this experiment,the body still had compensative capacity to maintain the balance of antioxidant system, (ii) Acrylonitrile exposure could injure the liver function at the 7th day, and then gradually returned to basal level after 7d.
