Objective: The topic of this experiment is to evaluate the plague subunit vaccine effect which is result from F1 +rV antigen in mice, guinea pigs two animal models , lay the foundation for the development of plague vaccine.Methods: 1. In mice animal models , this issue have used the mixture with the F1 , rV and aluminum hydroxide as vaccine . What is detected by splenic lymphocyte proliferation experiments, splenocyte cytokines, flow cytometry technique and the determination of antibody titers four methods . 2.In order to explore the appropriate concentration of the vaccine in the guinea pig animal model, experiment immune guinea pigs with different doses of F1 and rV antigen and aluminum hydroxide adjuvant . The immunogenicity of the vaccine were evaluated by separating the blood lymphocyte proliferation test and the determination of antibody titers two methods . 3. In effectiveness of vaccine protection experiments, Yersinia pestis attacks on the mouse, after a week with using different doses of the vaccine .The spleen bacteria were cultured to observe the protective efficacy of the vaccine.Results: 1.The mouse model experiment showed that ELISA has detected the F1+rV+Al(OH)3 group stimulating a strong humoral immune response; ELISPOT, MTT cell proliferation resμlts showed that the secretion of IFN-r in the spleen lymphocytes of F1+rV+Al(OH)3 group was higher than other groups at different stage. 2.The guinea pig animal model experiment showed that ELISA has detected the vaccine group stimulating a strong humoral immune response, and viable group of F1 antigen-specific humoral IgG titers significantly higher than the other vaccine group, the results were statistically significant. MTT cell proliferation results showed that it is no difference between the vaccine group and the negative control group at different time points after F1, rV antigen stimulated secretion of IFN-r in the spleen lymphocytes. 3.Experimental results show the effectiveness of protection of the vaccine negative control group, ranging from the number of colony growth, while the vaccine group were 0 spleen colonies, but the overall number of colonies were all below normal.Conclusion: The plague two-component vaccine in mice and guinea pigs can cause significant immune response, Al (OH) 3 adjuvant can significantly enhance the level of their response, and it has protect mice against plague.The experiment preliminarily confirmed that F1+rV+Al(OH)3 could stimulate a high level of immune response, and make a basis of the further development of new plague vaccines .
