| ObjectiveThe purpose of this study was to detect the expression of visfatinin serum of patients with preeclampsia(PE), and The -1535 promoter variant of the visfatin gene is associated with preeclampsia.MethodsA total of 96 women with PE and 96 normal pregnant women as control participated in this study.All are admitted to my hospital for delivery from July 2010 to January 2011.Patients with PE were divided into severe PE group(n=61) and mild PE group (n=35). Enzyme-linked immunosorbent assay(ELISA) was used to detect the levels of visfatin. Radioimmunoassay assay was used to detect the levels of fasting insulin. Homeostasis model assessment insulin resistance index (HOMA-IR). Multiplex SNaPshotSNP typing method used (single base extension) assay maternal visfatin gene promoter-1535C/T, determine the genotypes. Furthermore,clinical and biochemical parameters were recorded,such as body mass index(BMI),systolic pressure(SP),diastolic pressure(DP),total cholesterol(TC),triglyeride(TG),low density lipoprotein cholesterol(LDL),density lipoprotein cholesterol(HDL-C),apolipoprotein B(ApoB),apolipoprotein A1(ApoA1),fasting plasma glucose (FPG)Results①The level of serum visfatin was (62.30±23.41ng/ml) in PE groups, significantly elevated as(26.61±11.35ng/ml) in control group, the level in severe PE group was (67.08±26.25ng/ml) significantly elevated as(53.97±14.27ng/ml) in mild PE group.②The levels of TC, TG, LDL, ApoB, FINS and HOMA-IR, HDL-C and ApoA1 in PE groups significantly elevated as in control group. ③In PE groups, the serum visfatin was positively related to the serum TG, SP, DP, and HOMA-IR, while the serum visfatin was negatively related to the serum HDL-C. In control group, the serum visfatin was positively related to the serum TG, and HOMA-IR, and there was no correlation with SP and DP.④The distribution of visfation of genetype and allele frequencies of the SNP-1535 were different in obesity and normal control groups (P<0.05)。⑤In PE group,the levels of TG,HDL-C and serum visfatin were significantly elevated in the subjects with SNP-1535CC genotype as compared to TT genotype.In control group, the levels of TG and HDL-C were significantly elevated in the subjects with SNP-1535CC genotype as compared to TT genotype.⑥Logistic analysis proved that the SNP-1535CC genotype as compared to TT genotype in PE group was associated with increased risk of serum adiponectin level (OR= 1.026,95% CI:1.006-1.047, P<0.05). In PE group, visfatin gene promoter-1535CC-type patients, serum visfatin levels of the independent risk factors.Conclusions①Increased visfatin expression in serum be involved in the pathogenesis and development of preeclampsia.②Dyslipidemia and insulin resistance may be associated with increased visfatin expression in patients with preeclampsia, and may be involved in the pathophysiology of high visfatin hyperlipidemia led to development of preeclampsia③The-1535 promoter variant of the visfatin gene is associated with serum lipid.④The-1535 promoter variant of the visfatin gene is associated with the expression of serum visfatin⑤The-1535 promoter variant of the visfatin gene is associated with preeclampsia... |
