This project aims to the theory explore and experiment research of multi-component Pharmacokinetc-Pharmacodynamic Model, the Rhubarb Free Anthraquinones was used as the experimental subject, we detected both the Pharmacokinetic parameters and Pharmacodynamic of Rhubarb Free Anthraquinones on the treatment of intestinal injury in Beagle dogs, which was induced by severe acute pancreatitis, then we established the pharmacokinetic-pharmacodynamic model based on semi-mechanistic. SEM was used to describe the pharmacodynamic data, then first order ordinary differential dynamical systems of indirect-response PD model was established, after using three cubic spline interpolation smooth the PD date of SAP dogs, Runge-Kutta method was adopted to calculate the PD model parameters; after the PD mode was established, we used maximum effect model, linear model and zero-order model to connect multi-component Pharmacokinetc-Pharmacodynamic Model, evaluated the effect intensity of each component by goodness of fit and AIC, estimated the PD parameters of each drug. In order to accomplish the tasks mentioned above, the project evaluated the absorption kinetics parameters of the drug, dynamically evaluated the intestinal injury animal model on dogs induced by severe acute pancrestitis. All above, this project achieved the research aims in exploring multi-component Pharmacokinetc-Pharmacodynamic Model.
