Polygonatum odoratum mainly distributed in Jilin, Heilongjiang, Liaoning, Hunan, Neimenggu, Hebei, Gansu, Jiangxi, Shanxi, and Qinghai province. It was rich in resources. Summing up related Literatures; we found it was fewer about fresh Polygonatum odoratum in aspect of chemical constituent and antioxidation research of ethanol extract, which limited development and utilization of these resources. In this paper, chemical constituent and components of antioxidant activity of fresh Polygonatum odoratum were system studied, for further development of Liliaceae medicinal plants provide theoretical basis. The main conclusions as follows:(1) The DPPH radical clear activities were determined using DPPH method for fresh Polygonatum odoratum and dry Polygonatum odoratum aqueous two-phase extraction of petroleum ether, ethyl acetate, n-butanol, ethanol extracts four compositions . The results showed that dry and fresh Polygonatum odoratum root of the extraction of certain components have scavenging capacity to DPPH radical. When the same concentration of dry matter, each extraction component of fresh Polygonatum odoratum has lower IC50.Fresh Polygonatum odoratum root has the strongest scavenging capacity to DPPH radical, then it has the strongest antioxidant activity.(2) The chemical constituent of the 95% ethanol extract of fresh Polygonatum root were separated and purificated using silicage and Sephadex LH-20 chromatographic techniques.Eight compounds were isolated from the extracts of ethyl acetate and n-butanol fraction, through the identification of physical and chemical properties and spectroscopic,the compounds were respectively:3-O-β-D-glucopyranosyl-(l→2)-[β-D-xylopyranosyl-(l→3)]-β-D-glu-copyranosyl-(l→4)-galactopyranosyl-25(S)-spirost-5-en-3β-ol(G1);3β,26-diol-25(R)-Δ5,20(22)-dienofurostan-26-O-β-D-glucopyranoside(G2);palmitic acid(G3);stigmasterol(G4);daucosterol(G5) and three Fatty acids. Compound G2[3β,26-diol-25(R)-Δ5,20(22)-dienofurostan-26-O-β-D-glucopyranoside]was isolated from Polygonatum odoratum for the first time.The innovation of the paper:(1) Screening antioxidant active ones of each extraction fractions of Polygonatum odoratum with DPPH method, and identified the antioxidant active fractions.(2) The chemical constituent of the 95% ethanol extract of fresh Polygonatum odoratum root were separated and purificated using silicage and Sephadex LH-20 chromatographic techniques.
