Objective: To study the optimal activities and the role of curing the liver cancer for 125I seeds interstitial brachytherapy by investigating the therapeutic effect and pathological changes of rabbits VX2 liver tumor treated with different activities of 125I seeds combined with TACE.Methods:The New Zealand white rabbits ,body weight 2.5-3.0kg,male were provided by experiment animal center of Nanjing First Hospital affiliated to Nanjing Medical University. 96 cases rabbits of VX2 liver transplantation for hepatocellular carcinoma were randomly divided into 4 groups.:①the control group(0mCi125I seeds)( Group A,12 cases);②single 125I seeds group(group B,36 cases):1.0mCi 125I seeds(group B1,12 cases),0.7mCi 125I seeds(group B2,12 cases),0.4mCi 125I seeds(group B3,12 cases);③125I seeds +TACE group(group C,36cases):1.0mCi125I seeds+TACE(group C1,12 cases),0.7mCi125I seeds+TACE(group C2,12 cases),0.4mCi125I seeds+TACE(group C3,12 cases).;④TACE group(group D,12 cases).According to treatment planning system(TPS),group A,B,C were implanted different activity seeds by B–ultrasound;Group C,D were given TACE by DSA.We set four points:prior-treatment point,2weeks ,4 weeks ,6 weeks after treatment. The tumor volume was observed in order to calculate the tumor inhibitory rate. The tumor and surrounding normal tissue histopathological examinations,Blood parameters were performed.Results: Tumor size in every groups were not statistically significant difference before treatment(P >0.01). In every treatment groups( group B -group D ) , tumor size were statistically significant difference at the different time between prior-treatment and post-treatment (P <0.01).However ,they were smaller than in control group at the same time (group A) (P <0.01). The tumor inhibitory rate between prior-treatment and post-treatment were statistically significant difference by oneself(P <0.05) .The tumor inhibitory rate between the treatment groups had a significant change in two weeks after given the treatment(P<0.01), but all rate of treatment groups were higher than the rate of group D. There were statistically significant difference between group B1 and B2 in the tumor inhibitory rate.There are also the same between group C1 and C2.They were higher than the rate of group B3,C3,D (P<0.01).Pathological examinations(6 weeks after treatment) showed small amount of tumor cells were still observed in some tissues of group B3,C3,D.It could not be seen in others. The injury happened in normal liver tissue in group B1 was more serious than that in others. Gourp C2 were more suitable.The radiant hepatic injury were more serious in group B than the others,especially group B1.The efficacy in Group C2 were superior to that in others .Conclusion: 125I radioactive seeds implantation combined with TACE for the treatment of Liver cancer can block the blood supply and cure by chemotherapy on one side,the ray released by 125I radioactive seeds can kill the tumor cells and inhibit the tumor regeneration on the other side.Then,they were superior to a single treatment .Therefore,it is the safe,effective,suitable treatment for liver cancer.0.7 mCi would be the optimal activity for the treatment of Liver cancer.
