ObjectiveThe effects of PAF antagonist safflor yellow on heart function were investigated based on the observation of cardiac function, Left ventricular weight, left ventricular weight index and IL-1βmRNA expression of cardiac muscle by PAF antagonist safflor yellow treating the rats after acute myocardial infarction.Methods54 male SD rats were randomly divided into three groups: 18 rats in sham group (SH group), 18 rats in myocardial infarction group (MI group) and 18 rats in safflor yellow treated group (SY group). Acute myocardial infarcted animal models were established by ligation of anterior descending coronary artery. The rats in SY group were intervened by injecting intraperitoneally with 40mg.kg-1.d-1 safflor yellow after 24 hours, and the rats in MI group and SH group were injected with equal volume of saline. The heart functional indices were detected by Vivid 7-type Doppler ultrasound after 28 days, the left ventricular weight were measured and left ventricular weight index were calculated. And the IL-1βmRNA expression of cardiac muscle was detected by RT-PCR.Results1. The area of myocardial infarction in MI group and SY group was pale, left ventricular cavity was enlarged, the posterior ventricular wall became thick and the anterior ventricular wall obviously became thin.2. Compared with SH group, the thickness of left posterior ventricular wall, and the diameter of left ventricle in end-diastole and in end-systole were increased obviously in MI group and SY group. The left ventricular ejection fraction was decreased (P < 0.05), but the cardiac indices in SY group were better than that in MI group (P < 0.05).3. Compared with SH group, the left ventricular weight and left ventricular weight index were increased obviously in MI group and SY group, but the indices in SY group were improved(P < 0.05).4. Compared with SH group, IL-1βmRNA expression of cardiac muscle in MI group and SY group were increased significantly (P<0.05), but the IL-1βmRNA expression in SY group was less than that in MI group(P<0.05).Conclusion1. The heart function is reduced after acute myocardial infarction.2. PAF antagonist Safflor yellow could improve the heart function in rats after acute myocardial infarction.3. Platelet activating factor antagonist Safflor yellow probably improve the heart function by inhibiting inflammatory cytokine IL-1βmRNA expression of cardiac muscle after acute myocardial infarction in rats.PART II EFFECTS OF PAF ANTAGONIST SAFFLOR YELLOW ON TYPE-I,TYPE-III COLLAGEN AND MMP9MRNA EXPRESSION AFTER ACUTE MYOCARDIAL INFARCTION IN RATSObjectiveThe effects and possible mechanisms of PAF antagonist safflor yellow on left ventricular remodeling were investigated based on the Content of type-I collagen, type-III collagen, I/III collagen ratio and MMP9 mRNA expression level in non-infarcted myocardial area in rats after acute myocardial infarction.Methods54 male SD rats were randomly divided into three groups: 18 rats in sham group (SH group), 18 rats in myocardial infarction group (MI group) and 18 rats in safflor yellow treated group (SY group). The acute myocardial infarcted animal models were established by ligation of anterior descending coronary artery. The rats in SY group were intervened by injecting intraperitoneally with 40mg.kg-1.d-1 safflor yellow after 24h, and the rats in MI group and SH group were injected with equal volume of saline. Type-I and type III collagen were observed under the polarized light microscope after picric acid Sirius red staining, the volume fraction of collagen and I / III ratio were calculated, and MMP-9mRNA expression was detected by RT-PCR.Results1. Type-I and type III collagen in MI group and SY group were significantly increased, but those were improved in SY group(P <0.05). The ratio of I/III collagen among the three groups was statistically significant (P <0.05).2. The MMP-9mRNA expression was increased in MI group and SY group (P <0.05), but it was better in SY group than that in MI group (P <0.05).Conclusion1. PAF antagonist safflor yellow could improve left ventricular remodeling after acute myocardial infarction in rats.2. The effects maybe related to the following aspects:(1) PAF antagonist safflor yellow could improve the expansibility of cardiac tissue by reduce type-I/III collagen and I / III collagen ratio.(2) PAF antagonist safflor yellow could reduce MMP-9 expression and activity, and it could reduce the collagen deposition.
