Effects Of Mitiglinide Calcium On Fasting Plasma Fibroblast Growth Factor-21 Levels In Patients With Type 2 Diabetes Mellitus

Objective:Fibroblast growth factor-21 (FGF-21) has recently been characterized as a glucose and lipid metabolism regulator. This study aimed to investigate the effects of Mitiglinide Calcium on plasma FGF-21 levels in patients with type 2 diabetes mellitus (T2DM).Methods:This is a case-control study. Eighty-two patients with new-onset T2DM and 82 sex- and age-matched normal glycaemic controls (NGT) were enrolled in the study. The T2DM group was treated with Mitiglinide Calcium for 16 weeks. Plasma FGF-21 levels were measured by ELISA in patients with T2DM and NGT. FGF-21 levels between pre- and post-treatment with Mitiglinide Calcium were compared and the relationship between plasma FGF-21 levels and BMI,WHR, HOMA-IR and other metabolic parameters were also analyzed.Results:Fasting plasma FGF-21 levels were higher in T2DM group than that in the control (2.57±0.54 vs 1.84±0.54μg/L,P<0.01), Plasma FGF-21 level was positively correlated with TG and FINS (r=0.32, r=0.41, P<0.05 or P<0.01), while negatively with HbA1c and TC (r=-0.25, r=-0.24, both P<0.05). In T2DM patients, multiple regression analysis showed that HbA1c and HOMA-IR were associated with plasma FGF-21 levels. Compared with pre-treatment, fasting plasma FGF-21 levels were decreased significantly in T2DM group after treatment with Mitiglinide Calcium (2.57±0.54 vs 2.14±0.78μg/L,P<0.01), and the reduced FGF-21 were correlated with reduction of FINS and HOMA-IR.Conclusions:Plasma FGF-21 level was increased in T2DM patients, but significantly decreased after treatment with Mitiglinide Calcium. As a newly anti-diabetic agent, Mitiglinide Calcium may play a role in regulating plasma FGF-21 level in diabetes patients.
