Objective: To detect the expression of the novel ATRA-related resistance gene HA117 in lung cancer and breast cancer. The resistance mechanisms related to the gene HA117 were studied,which were beneficial for the chemotherapy choosing and prognosis evaluation.Methods: 23 specimens of lung cancer and 21 specimens of breast cancer were randomly selected as experimental group, while 9 and 11 specimens of non-cancerous tissue of lung and breast as control group correspondingly. Streptavidin-peroxidase immunohistochemistry(IC) staining method was employed to detect the expression of HA117 in all specimens above and retrospective analysis was conducted combining their clinical data.Results:1. In the specimens of lung cancer, the positive expression rate of the gene HA117 was 60.9%(14/23). Meanwhile, only 11.1%(1/9) was detected in the specimens of non-cancerous lung tissue correspondingly. The data indicated that the positive expression of the novel gene was significantly different between lung cancer and non-cancerous lung tissue (p<0.05).2. The positive expression of the gene HA117 showed no significant correlation with age of patients, tumor grades, pathological types, lymph node metastasis in the cases with lung cancer(all p>0.05).3. In the specimens of breast cancer, the positive expression rate of the gene HA117 was 57.1%(12/21). Meanwhile, no positive expression was detected in non-cancerous breast tissue correspondingly. The data indicated that the positive expression of the novel gene was significantly different between breast cancer and non-cancerous breast tissue (p<0.05).4. The positive expression of the gene HA117 showed no significant correlation with age of patients, pathological grading, depth of local invasion, lymphnode metastasis and preoperative chemotherapy in the cases with breast cancer (all p>0.05).Conclusions:1. The high expression of the gene HA117 in the specimens of lung cancer indicated presence of strong resistance to ATRA which has no relationship with patient's age, pathological grading, pathological types and lymphnode metastasis.2. As well as lung cancer tissue,the breast cancer tissue had a high expression of the novel gene.Which indicated that strong resistance to ATRA still exists in breast cancer tissue with the same correlation with patient's age, pathological grading, depth of local invasion, lymphnode metastasis and preoperative chemotherapy.3. The strong resistance to ATRA in two kind of cancer provided theoretical basis for chemotherapeutic agents choosing.The resistance strength of patients must be assessmented and reversed by using ancillary drugs.In order to improve the effect of chemotherapy,individual treatment plan should be made soon afterwards in clinical treatment.
