ObjectiveTransfect Survivin siRNA to colon cancer SW480 cell lines and study interfe-rence on proliferation and apoptosis to provide the gene therapy for colon cancer experimental basis and seek new,effective RNA interference fragmentMethodsThree siRNA targeting survivin genes were constructed in vivo and transfected into SW480.At 24h,48h,72h, the change of survivin protein was detected by Westem blot,at the same time,cell proliferation was dectected by MTT assay and Flow cytometry.ResultsThree siRNA effected on SW480 cells at 24h,48h,72h and three groups appeared that Survivin protein expression decreased (P<0.05).The expression of survivin protien reached apex at 48h and the inhibitory rates were 66.5%±2.1%,49.6%±2.8%and 47.8%±3.1%, respectively, compaired with the control group (P<0.05); MTT assay showed cell proliferation was inhibited after survivin gene silencing;Flow cytometry showed SW480 had obvious changes in cell cycle:Go/G1 phase cells increased and G2/M phase decreased after transfection.Conclusion 1. Liposome-mediated survivin-siRNA can effectively silence the survivin gene in colon cancer cells, inhibit cancer cell proliferation and promote apoptosis.2. especially designed for targeting conserved regions of siRNA, interference effects is more significant.3. The results provide a good interference sequence for the next tumor model in nude mice.
