Objective To assess the expression of PKCβ/CPI-17 system in uterus smooth muscle and elucidate its role of onset of labor.Method 15cases of term pregnancy without labor (NL group), 15case of term pregnancy with labor(L group) and 15case of nonpregnant women (NP group) undergoing hystereetomy due to cervical intraepithelial neoplasia were recruited into this clinical study. Real-time quantitative polymerase chain reaction and Western-Blot method was used to evaluate the expression of PKCβand CPI-17 mRNA and protein in myometrium.Result 1.The expression of PKCβmRNA in L group was significantly higher than that in NL group, P<0.01, which was significantly higher than that in NP group, P<0.01; 2. There was not significantly difference among the expression of CPI-17mRNA in L, NL and NP group,P>0.05; 3.The expression of PKCP protein in L group(0.56±0.20) was significantly higher than that in NL (0.43±0.13)group, P<0.05, which was significantly higher than that in NP group(0.19±0.01),P<0.01; 4.The expression of CPI-17 protein in L, NL and NP group were (0.50±0.31), (0.48±0.35), (0.45±0.32), there was not significantly difference among L, NL and NP group, P>0.05; 5.The expression of phosphorylated CPI-17 protein in L group(0.65±0.19)was significantly higher than that in NL group(0.43±0.22), P<0.05, which was significantly higher than that in NP group (0.31±0.02), P<0.05; 6.Positive rank correlation between the expression of PKCβand phosphorylated CPI-17 protein in Pregnancy uterus smooth muscle was observed (r=0.84, P<0.01).Conclusions The expression and activity of PKCβ/CPI-17 in the uterine smooth muscle may play an important role in the onset of labor.
