Objective:To study the potential function of miR-140 in develop-ment and progression of Osteoarthritis (OA) using real-time PCR.Materials and methods:The expression level of miR-140 in knee synovial fluid was detected by real-time PCR (stem-loop RT-qPCR, TaqMan). Knee synovial fluid specimens were divided into two groups: 1,the group of OA patients 40 cases(According to X-ray diagnosis criteria of Kellgren and Lawrence:gradeⅠ(10 cases),gradeⅡ(10 cases),gradeⅢ(10 cases),gradeⅣ(10cases);2,the control group(20 cases)(meniscus injury 10 cases,cruciate ligament injury 5 cases,trauma 5 cases) patients. The expression levels of miR-140 in knee synovial fluid were detected by qPCR. The relationship between the severity of osteoarthritis and the expression levels of miR-140 was analysed. U6 snRNA was used as endogenous control.All data was analyzed by SPSS 17.0 statistical software.α=0.05 was regarded as the statistical criterion.Results:1. MicR-140 can be detected in all knee synovial fluid.2. The expression level of miR-140 in OA patients is much more lower compared with control group.(P<0.05).3. The expression levels of miR-140 and the severity of OA was negatively correlated.r=-0.917. Conclusion:1. There is miR-140 in human knee synovial fluid, which can be detected conveniently by real-time PCR.2. The expression level of miR-140 is lower in the knee synovial fluid of OA than control group,and have great correlation with the severity and progression of osteoarthritis(OA).
