Objective:To study the relationship between hOGG1, XRCC1, XRCC3 gene polymorphisms and the effect of radiotherapy and prognosis in esophageal squamous cell carcinoma. Methods:Phlebotomize 199 patients of esophageal squamous cell carcinoma and extract genome DNA from whole blood cells, hOGG1, XRCC1, XRCC3 genotypes were determined by PCR-RFLP method. Analyse the relationship between the SNP of hOGG1, XRCC1, XRCC3 and the Effect of Radiotherapy and Prognosis in ESCC according to clinical and follow-up data. Result:199 esophageal squamous cell carcinoma patients who were treated with radical radiotherapy got the total response rate is 81.4%. The response rate to radiotherapy in patients with the Thr/Met genotype was significantly higher than that in patients with Thr/Thr genotype (P=0.017). The incidence of esophageal radioactivity injury in patients with the Thr/Met genotype was also significantly higher than that in patients with Thr/Thr genotype (P=0.020). The radiotherapeutic recent effect in patients with DT2;60Gy was better than that with DT<60Gy(81.4%vs61.3%, P< 0.001), while it would not increase the incidence of esophageal radioactivity injury in patients with DT≥60Gy (25.2%vs23.2%, P>0.05). The median survival period in patients with Thr/Thr genotype and Thr/Met genotype was 14 months and 27months respectively, which were statistically significant(P=0.001). According to multivariate analysis, the SNP of XRCC3 and clinical stage,which were closely related to over all survival,were the independent prognostic factors (P=0.029,0.001). Conclusion:1.The radiosensitivity of XRCC3 Thr/Thr genotype was better than Thr/Met genotype.2.The radiotherapy efficacy with DT≥60Gy was better than that with DT<60Gy, while it would not increase esophageal radioactivity injury.3.The SNP of XRCC3 and clinical stage were closely correlated with prognosis. XRCC3 Thr/Thr genotype and higher clinical stage were unfavorable factors for the prognosis of esophageal squamous cell carcinoma.
