Objective In order to study mechanism of lymphangiogenesis of colorectal carcinoma(CRC), Research in colorectal cancer organizations hypoxia-inducible-1 alpha(HIF-1α),von Hippel-Lindua(VHL)expression.Discusses their generation and lymphatic in colorectal cancer invasion and metastasis, further studies the role of generating regulation mechanism lymphatics.Methods 60 patients with colorectal cancer patients review of its medical records, the cancerous tissue and the far cancer cut colon tissue specimens with fluorescent real-time quantitatie polymerase chain reaction (PCR) method detection VHL hypoxia-inducible-1 alpha, the mRNA transcription levels, while using immunohistochemistry on biopsy dyed detection VHL hypoxia-inducible-1 alpha, the protein expression circumstance, to fight podoplanin monoclonal antibody mark determination micro lymphatic density, analysis VHL hypoxia-inducible-1 alpha, molecular mRNA transcription and protein expression level and clinical pathologic material, between the relationship between micro lymphatic density.Results The levels of HIF-1αof tumor tissues were higher than that of surigical margin in either mRNA transcription or protein expression. The levels of VHL of tumor tissues were lower than that of surigical margin in either mRNA transcription or protein expression.A significant negative relation between VHL expression and HIF-1αwas showed.There were a statistics difference in the differentiation of tumor, the depth of tumor,lymphatic node metastasis as well as Dukes stage of colorectal cancer in both mRNA and protein of HIF-la and VHL.The different expresses of them in different age,gender,primary tumor location and pathologic type were not detected.Conclusions There is richer formation of lymphatic vessels in CRC than surgical marginal tissue. The HIF-1αand VHL are important positive regulator of lymphangiogenesis, could be used in judgment of metastasis and prognosis of CRC.
