Objective:To investigate the expression pattern of IGF-1 in colorectal cancer and its relationship with lymphangiogenesis and lymphatic metastasis of colorectal cancer.Methods:Selected were confirmed by pathology,surgical resection of colorectal cancer samples from 40 cases of tumor and adjacent normal tissues,each sample was divided into 2 parts,one is placed in 10%formalin,parafin section and immunohistochemical analysis,another part for the extraction of miRNA.Analysis of IGF-1 expression patterns and their relationships in their respective organizations.Results:The expression of IGF-I/IGF-I R in the adjacent normal tissues was 1.40±0.59,1.30±1.18,in the tumor tissues were 4.53±2.50,4.65±2.66,IGF-I/IGF-I R group was statistically significant(P<0.05).The expression of IGF-I/IGF-I R in different differentiation tissues,high differentiation group were 3.0±1.41,2.11±0.93,moderate differentiation groups were 4.30 ±2.64,4.55±2.35,low differentiation group were 6.18± 2.09,6.64±2.11,there were statistically significant differences between the groups(P<0.05).IGF-I/IGF-I R expression in no lymph node metastasis tissues were 3.12±1.54,3.52±2.18,respectively 6.87±1.99,6.33±2.23 in lymph node metastasis expression in tissues,the two groups were statistically significant difference(P<0.05).The count is 11.45±3.03 lymphatic vessels in colorectal cancer tissues,paracancerous tissue was 3.25±1.28,two groups had significant difference(P<0.05),lymph node metastasis lymph tumor tissue tube count was 12.67±4.09,no lymph node metastasis tumor lymph vessel counts was 10.72±1.90,two groups were significant difference(P<0.05).In the migration and invasion experiments,the number of LoVo cells in the IGF1 treated group was(196.4± 21.6),and the number of penetrating membrane was(163.6 ±19.4),which was significantly higher than that of the control group(96.4 ±11.2),and(72.5±9.1),the difference was statistically significant(P<0.01).IGF1 on the growth of transplanted tumor in nude mice,IGF1 treatment group tumor volume and weight were 983.6 ±124.7 and 2.2 ± 0.6,the control group was 781.4±97.1,1.8±0.6,the difference between the two groups was statistically significant(P<0.05).IGF1 on the transplanted tumor LVD in nude mice,IGF1 treatment lymphatic count was 10.7±3.3,physiological saline group was 6.4 ± 2.9,the difference between the two groups was statistically significant(P<0.05).Conclusions:1.In colorectal cancer,IGF1/IGF1R can promote lymphatic metastasis by inducing lymphangiogenesis.2.There was a negative feedback regulation relationship between IGF1/IGF1R and let-7 miRNA,and the interaction between let-7 and IGF1/IGF1R indirectly affected the formation of Lymph.The negative feedback regulation relationship between IGF1/IGF1R and let-7-miRNA may provide new clues for clinical anti-tumor lymphangiogenesis.3.IGF-I/IGF-I R plays an important role in tumorigenesis and development,and has important value in colorectal cancer diagnosis,prognosis and treatment decisions.
