Age-related cataract is the most common type of cataract. At present, there are many researches scholars think that age-related cataract is protein structure change by lens, lens protein disease caused the irreversible degeneration and lens the protease abnormal decomposition relevant. In this paper the proteins and enzymes from the lens altered actiity Angle discusses the age-related cataract causes of disease.20s Proteasome with hydrolysis normal and abnormal protein function, in the cell cycle, apoptosis, signaling physiological activities play an important regulatory role. It has peptide respectively carbonyl glutamine peptide (PGPH) hydrolysis reactions, trypsin samples (Tom lantos) hydrolysis reactions and mi protease sample (CT-L) hydrolysis reactions. Already proof in a variety of cells and tissues, the aging process 20s Proteasome activity, content and the expression level change, these changes are highly cells and tissues specificity. The Proteasome 20s in people's lens of age-related cataract may also have a similar effect, but has yet to see related reports.Objective:to study the 20s Proteasome with age-related cataract onset relationship, this experiment of age-related cataract selected lens for research object, detection in the 20s Proteasome lens express changes cataract.Methods:through Western-Blot method to detect the 20s Proteasome in the content of age-related cataract lens changes, and through the fluorescent marker protein substrates method to detect changes in the 20s Proteasome enzyme activity.Results:this experiment through Western-Blot method to detect in people the age-related cataract 20s Proteasome content in the lens, the results show that, with the opacity degree and the nuclear hardness lens increased,20s Proteasome content tended to decline trend; Enzyme activity determination results show that, with the opacity degree and the nuclear hardness lens increased,20s Proteasome enzyme activity also assumes a downward trend.Conclusion:the results show that, as age-related cataract lens cloudy degree and the nuclear hardness increases,20s Proteasome content declined, 20s Proteasome of the enzyme activity also shows trend to decrease. Thus we speculate which may result in the development of some lens with lens on the change, indicating protein 20s Proteasome content in cataract lens of change, and cataract disease related.
