Objective: By detecting the expression level of 20 s proteasome in lens anterior capsule and nucleus with the lens nucleus hardness changed, to explore the interconnections between the degradation of apoptosis factor and oxidized protein on 20 s proteasome and age-related cataract happening.Methods: Chose the patients who were diagnosed as age-related cataract from January 2014-2014 in July at the Wannan Medical College affiliated Yijishan hospital wards of Department of Ophthalmology, except with the other eye diseases and systemic diseases.30 cases of cataract lens nucleus were collected in small incision extracapsular cataract extraction. According to the nuclear hardness, they were divided into three groups, including A(II soft core), B(III medium hardness nuclear), C(IV hard core, V very hard core). Compared sex, age, eye, period in three groups of lens nucleus. After it showed there were no difference on them, applied the Western blotting method to detect the content of 20 s proteasome in the three groups of age related cataract lens nucleus; 30 cases of cataract lens anterior capsule were collected in small incision extracapsular cataract extraction. According to the nuclear hardness, they were divided into two groups, including A( II soft core), B(IV hard core, V very hard core).Compared sex, age, eye, period in three groups of lens anterior capsule.After it showed there were no difference on them, applied the Western blotting method to detect the content of 20 s proteasome in the two groups of age related cataract lens anterior capsule. SPSS17.0 statistical software was used for statistical analysis of the date.Results: It was no statistically significant difference of gender, age, eye, the onset time among the three groups in the lens nucleus(P > 0.05); And it was also no statistically significant difference of sex, age, eye, the onset time between two groups in lens anterior capsule(P > 0.05). Western blotting method was used to detect the content of 20 s proteasome in the three groups,it showed that difference among the three groups in the lens nucleus was statistically significant(P <0.01); The content of 20 s proteasome in soft core group were higher than hard core、very hard core group of lens anterior capsule sample.Conclusion: With the opacity degree and the nuclear hardness lens increased, 20 s proteasome content of age-related cataract lens nucleus and anterior capsule tended to decline. While 20 s proteasome is an important enzyme which remove apoptosis factor and oxidation protein.The reduce of 20 s proteasome content relates the abnormal changes of lens protein, it indicates the change of 20 s proteasome content is closely related to the occurrence and development of age-related cataract.
