| Raman spectrum, which is characterized by the frequency shift of molecular vibration or rotation energy level difference that caused by interactions of molecule and photon, is a kind of molecule scattering spectroscopy. Different material or molecule has different fingerprint Raman spectrum. Therefore, it can be used to analysis and detect the structure or component of other substance. Due to its characteristic of non-osculatory, non-destructive, high sensitivity, short time, a spot of sample and non-preparative sample, Raman spectrum, which made extraordinary contribution to the social development and progress, are widely used in material, environment, chemistry, biomedicine, archeology etc.This thesis introduced the theoretical knowledge and application as well as data analysis and processing with Raman spectrum. Meanwhile the data processing method was applied for the research in diagnosis of serum of breast cancer and healthy. The main contents of this thesis are as follows:(1) Raman scattering effect is explained from the viewpoint of theory of quantum and classical respectively as well as two kinds of enhancement mechanism:physical enhancement and chemical enhancement. In addition, the Renishaw's structure of dispersion types raman spectrometer and the advantages of raman spectrum as well as different application area is also described.(2) In order to reduce plenty of high-frequency random noise, fluorescence background, light scattering noise result from nonuniformity of sample in the signal of raman spectrum, different data processing is applied to reduce the noise and improve signal to noise ratio.(3) SERS of serum from breast cancer patients and healthy volunteers is analyzed in molecular level. The changes of each amount of components contained in different substance, molecular structure and functional group are found. Besides, the difference of serum in canceration and healthy is pointed out. And the database of serum of SERS from breast cancer patients is established, which will provide the references for clinical diagnosis and treatment.In this thesis, two different methods PCA and LDA are adopted so that we can distinguish between the serum of breast cancer patient and healthy volunteer. Consequently, a satisfactory result, with which successfully achieved the goal of clinical diagnosis in the sight of raman spectrum, was obtained. |
