Objective:After the traumatic stimulus at the stomach of the rats, the convergence and integration of information afferents at electro-acupuncture of Sibai (ST 2) and the traumatic sensory afferents on nucleus of tractus solitarii (NTS) and paratrigeminal nucleus (PTN) were observed. By exploring the specific roles of PTN in the characteristic relation between ST 2 and stomach, it provides the scientific basis for the correlativity of acupoints with zang-fu organs (ST 2 and stomach) and the scientific basis for the acupuncture analgesia.Method:using the triple-labeled methods of FG retrograde trace combined with Fos and Calbindin D-28k proteins immunofluomscenee histochemistry, Observe the retrogradely labeled neurons and the transganglioniely labeled fibers/terminals in NTS and Pa5 in electro-acupuncture of Sibai (ST 2) and the visceral pain rats.â‘ Neurons in Pa5 receive visceral noeiceptive information directly project to NTS:using the triple-labeled methods of FG retrograde trace combined with Fos and CB proteins immunofluomscenee histochemistry, first inject the FG into NTS as a model, then stimulating the stomach with formalin, Observe whether the labeled neurons with CB and FG exhibited positive Fosimmunoreaction.â‘¡Neurons in Pa5 receive information by EA ST2 directly project to NTS:using the triple-labeled methods of FG retrograde trace combined with Fos and CB proteins immunofluomscenee histochemistry, first inject the FG into NTS as a model, then EA ST2, Observe whether the labeled neurons with CB and FG exhibited positive Fosimmunoreaction.â‘¢The information by EA ST2 can inhibit that from visceral noeiceptivation. Inject the FG into NTS as a model, stimulating the stomach with formalin, and then EA ST2 for 20min, Observe the differences on triple-labeled neurons with other groups.Results:1. After inject FG to NTS (the right hand side) of the rats and perfuse formalin to the their stomach, totally 7 kinds of memory cells which are the single label of Fos, CB and FG, double labeled Fos/CB, Fos/FG, CB/FG and triple-labeled Fos/CB/FG appear in both side of PTN and NTS, chiefly on the right hand side. There are about 44.5 neurons single labeled with FG,56.5 with CB and 79 with FOS, among wich 22.83 neurons with CB/FG double label, occupies that with FG 51.31%,26.5 neurons with FOS/FG double label, occupies that with FG 59.55%,28.17 neurons with CB/FOS double label, occupies that with CB 49.85%,13.33 neurons with Fos/CB/FG triple-label, occupies that with CB/FG 47.37%.2. After inject FG to NTS (the right hand side) of the rats, and EA ST2 for 20min, the distributions of the labeled neurons are similar to the group above, on the right hand side there are 7 kinds of memory cells which are the single label of Fos, CB and FG, double labeled Fos/CB, Fos/FG, CB/FG and triple-labeled Fos/CB/FG. There are about 23 neurons single labeled with FG,28.83 with CB and 37.33 with FOS, among wich 14.17 neurons with CB/FG double label, occupies that with FG 61.60%,16.83 neurons with FOS/FG double label, occupies that with FG 73.19%,19.33 neurons with CB/FOS double label, occupies that with CB 67.05%,8.17 neurons with Fos/CB/FG triple-label, occupies that with CB/FG 42.24%.3. After inject FG to NTS (the right hand side) of the rats, stimulating the stomach with formalin for 2h, and then EA ST2 for 20min, the distributions of the labeled neurons are similar to the two groups above, on the right hand side there are 7 kinds of memory cells which are the single label of Fos, CB and FG, double labeled Fos/CB, Fos/FG, CB/FG and triple-labeled Fos/CB/FG. There are about 28 neurons single labeled with FG,36.67 with CB and 49.67 with FOS, among wich 18.17 neurons with CB/FG double label, occupies that with FG 64.88%,17 neurons with FOS/FG double label, occupies that with FG 60.71%,23.33 neurons with CB/FOS double label, occupies that with CB 63.64%,12.17 neurons with Fos/CB/FG triple-label, occupies that with CB/FG 49.67%.There are 49.67 FOS labeled neurons in Pa5 are less than the group of only stimulating the stomach with formalin (P<0.05).There was no significant difference comparing the group of only stimulating the stomach in the percentage of triple-labeled neurons to CB/FG double labeled neurons.Conclusions:EA ST2 can effectively ease pain from stimulating the stomach with formalin; The information afferents at electro-acupuncture of Sibai (ST 2) and the traumatic sensory afferents converge and integrate in Pa5 and project to NTS,wich play an important role in the therapy in stomachache. The neurons containing CB may integrate the information, wich is the key to therapy in stomachache through EA ST2.
