Objective:To investigate the expression and correlations of RUNX3,P21 and Cyclin D1 protein in human glioma,analyze their effect on the occurrence,development and the clinical pathological grade of human gliomas, for clarifying the pathogenesy of human glioma and providing new assumption for clinical treatment.Methods:58 cases of human glioma tissue were used to detect the expression of RUNX3,P21 and Cyclin D1 by streptavidin-perosidase(SP) immunohistochemical technique. According to WHO's tumor classification of central nervous system in 2000:Gradeâ… 6 cases,they are all astrocytoma:Gradeâ…¡18 cases,including 8 cases astrocytoma,7 cases oligodendroglioma,1 case pleomorphic xanthoastrocytoma and 2 cases ependymoma;Gradeâ…¢20 cases.including 11 cases anaplastic astrocytoma.8 cases anaplastic oligodendroglioma and 1 case anaplastic ependymoma;Gradeâ…£14 cases,including 9 cases glioblastoma and 5 cases medulloblastoma.Applying SPSS 17.0 statistical software to calculate the related data.Results:High expression rate of RUNX3,P21 and Cyclin D1 was 41.4%,48.3%,53.4% respectively in 58 cases of human gliomas.There was significant difference of RUNX3,P21 and Cyclin D1 among the different grades.There was significant difference of the RUNX3 expression between lower Grade(â… -â…¡) and higher Grade(â…¢-â…£),along with the pathological grade increasing of human gliomas,the expression level of RUNX3 decreased gradually(X2=38.624,P< 0.05),registering as negative correlation; There was also significant difference of the P21 expression between lower Grade(â… -â…¡) and higher Grade(â…¢-â…£),along with the pathological grade increasing of human gliomas,the expression level of P21 decreased gradually(X2=14.065,P <0.05),registering as negative correlation:There was significant difference of the Cyclin D1 expression between lower Grade(â… -â…¡) and higher Grade(â…¢-â…£),along with the pathological grade increasing of human gliomas,the expression level of Cyclin D1 increased gradually(X2=16.129,P<0.05),registering as positive correlation.Simultaneously.there was positive correlation between the expression of RUNX3 and P21 in human glioma,but negative correlation was discovered between the expression of RUNX3 and Cyclin D1.P21 and Cyclin D1.Moreover, the expression level of RUNX3,P21 and Cyclin D1 had no concern with gcndcr.agc.group of pathology typing.sizc and location of human glioma and chemoradiothcrapy. Conclusions:There was affinity between the expression level of RUNX3..P21,Cyclin D1 and pathological grading of human glioma. With the increase of glioma grade,RUNX3 and P21 protein expressions were decreased gradually. Cyclin D1 protein expression was increased gradually. There was negative correlation between the expression of RUNX3 and Cyclin D1,P21 and Cyclin D1. Moreover,there was positive correlation between the expression of RUNX3 and P21.We could gain the prompt that RUNX3 participate the malignant transformation of human glioma by effecting the expressions of P21 and Cyclin D1 protein.The effectiveness of synergism and rivalry between the three maybe had significance in hyperplasy and apoptosis of human glioma.
