Objective The studying detected the expression of RUNX3mRNA,RUNX3 protein in human cholangiocarcinoma tissue, and estimated theassociation of RUNX3 with cholangiocarcinoma and its clinicalsignificance.Methods Reverse transcription polymerase chain reaction (RT-PCR)was applied to detect the expression of RUNX3mRNA in 20 humancholangiocarcinoma tissues and cholangeitis tissues; immunohisto-chemistry was used to detect the expression of RUNX3 protein in 45cholangiocarcinoma tissues and 10 normal bile duct tissues.Results(1) Expression of RUNX3mRNA in cholangiocarcinoma tissue wassignificantly lower than that in cholangeitis tissues (p<0.01).(2) Expression of RUNX3 protein in cholangiocarcinoma tissue wassignificantly lower than that in normal bile duct tissue(p<0.01).(3) The expression of RUNX3 protein in cholangiocarcinoma tissuewas not related to the gender, age, and location of tumour (p>0.05).(4) The expression of RUNX3 protein was related to differentiation of cancer cell, metastasis of lymph node and clinical stage (p<0.01).Conclusions(1) Down-regulated expression of RUNX3 is obviously in humancholangiocarcinoma tissue.(2) Expression level of RUNX3 protein in human cholangioca-rcinoma tissue is not correlated with patients gender, age and location oftumour.(3) Expression level of RUNX3 protein in human cholangio-carcinoma tissue is correlated with differentiation degree of cancer cell,Clinical stage and metastasis of lymph node...
