Objective In this study, TanshinoneⅡA, the active ingredient of Tanshinone would be applied to huaman ovarian carcinoma cell SKOV3 in vitro. Observing its effects on proliferation and apoptosis of huaman ovarian carcinoma cell SKOV3 and the expression of survivin and smac/DIABLO and investigate its mechanism,and then provide some theory basis for its application for ovarian cancer.Methods①Human ovarian carcinoma cell SKOV3 was cultured in vitro and with TanshinoneⅡA at various concentrations and different intervention time. MTT assay wad used to detect the growth of SKOV3. The cell cycle and apoptosis rate was determined by flow cytometry.②The expression of survivin and smac/DIABLO mRNA at various concentrations and different intervention time were analyzed by RT-PCR.③The expression of survivin and smac/DIABLO protein were analyzed by fluorescence-immunocytochemistry.Results①TanshinoneⅡA could inhibit human ovarian carcinoma cell SKOV3 proliferation in the range of 1.0-10.0μg/ml,and it showed a time-dose-dependence. The experimental group had much more G1 phase cells and less S phase cells than the control group(P<0.05).②Survivin and Smac/DIABLO mRNA were both expressed in SKOV3 cell. The level of gene expression of Survivin remarkably was down-regulated (P<0.05), and the gene expression of Smac/DIABLO was remarkable up-regulated(P<0.05), and it showed a time-dose-dependence.③The protein of Survivin and Smac/DIABLO were both expressed in SKOV3 cell, most in cytoplasm and a few Survivin protein in nucleus.Conclusion TanshinoneⅡA can significantly inhibit the proliferation of human ovarian carcinoma cell SKOV3, change cell cycle and induce cells apoptosis, which related to down-regulate the expression of Survivin and up-regulate the expression of Smac/DIABLO in human ovarian carcinaoma cell SKOV3.
