| The herbal plant Acanthopanax Senticosus Harms (Araliaceae) is found widely spread in North China. Different parts of the plants have been used as ingredients of folk medicines for the treatment of a variety of human diseases. For Acanthopanax Senticosus Harms, Flavonoids occur in practically all parts of the plant and have the greatest content in leaves. The total flavonoids obtained from the aerial part of this plant have shown wide biological activities. In the article, the purification process of total flavonoids, UPLC fingerprints analyses were studied aiming to develop and take best advantage of Acanthopanax Senticosus.The main works and conclusions are included as follows:The purification process of total flavonoids in Acanthopanax Senticosus leaves with macroreticular resin were studied. The best technics of purification is as follows:the type of macroreticular resin is DM130, the consistency of eluting solution is 40% ethanol solution, the amount of water is 4-5 times as much as the amount of macroreticular resin.the amount of elution is 4-5 times as much as the amount of macroreticular resin and the speed of eluting is 4BV/h, then concentrate the fluid, dry it, the intermediate contain more than 60% total flavonoids. It is suited for the production and possess spacious prospect.UPLC analyses were carried out to get as many compounds as possible in these components. From the results, it shows the best technics as follows:Zorbax Agilent C18 (4.6 mm×150 mm 5 apms m) chromatographic column, column temperature:30℃, the consistency of eluting solution are acetonitrile and water, the proportion is 10:90, the speed of eluting is 0.8 ml/min, detected wavelength for 265nm.sign the chromatograms after the sample inject into the UPLC.The result show that the content of syringin still changes in different growth stages. UV were used to test 10 samples from two different cultivation methods. In conclusion, Human factors have influence on growth movement of the Acanthopanax Senticosus Harms. |
