Objective: To investigate the inhibitory effect of Didangqigui Soup on the occurrence and development of SD rats'diabetic cataract, and provide clinical reference for its prevention and treatment of diabetic cataract.Methods: Rats were randomly selected as control group (group A), with intraperitoneal injection amount citric acid buffer A, the other 70 rats were given chain meal (STZ 60mg/kg) to make diabetic model after fasting 12 hours,72 hours later measured their blood sugar .The blood sugar level which≥16.9mmol/L, urine sugar with more than +++~+++,were both regarded as made mould success. 14 rats were removed, the remaining 56 randomly divided into 5 groups respectively: model control group (group B), western medicine group (group C), Chinese traditional medicine normal dosage group (D group), Chinese traditional medicine high-dose group (group E), Chinese medicine with western medicine group (normal doses of group F).Then gave corresponding intervention. Then 0.5% tetramin methenamine amide eye drops are used as mydriatic in week 2, 4, 6 and 8.The condition of lens opacity were examined and recorded in the slit lamp. Eight weeks later ,blood glucose were determined before the determination of death by 10% hydrated chlorine aldehyde anesthesia fatal overdose. Then eyes were quickly removed, fixed by more than 4% after fixation gather formaldehyde and made frozen section as 0.4μm thick. After HE dyed,the change of the lens epithelial cells wer e observed under optical microcopy . And the expression of HSP70 in LEC was detected by immunohistochemical SP ldpe-g-nvp.Results: Rat lens of Group A always maintain transparent completely, rats lens of group B gradually opacity. Compared with group A, each group lens happened different degree of opacity, crystal determinated by heavy to light in turn for group E >group D≥group C≥group F >group A; HE dyeing lens epithelial cells form also change. The expression of HSP70 in LEC in group A was standard,which in group B increased gradually. That is higher than group C and D group. The expression of HSP70 in LEC in group F with significant difference (P < 0.05), group C, D group, the group F HSP70 positive expression rate between non-significance difference (P > 0.05).Conclusion: Didangqigui Soup could effectively protect the damage of experimental diabetic rats lens, inhibit the occurrence and developmen of diabetic rats catarac,especially in the early disease development. The mechanism may be that:it could lower blood sugar, reduce sugar metabolites gathered inside lens ,and lower osmotic pressure inside lens, so that lessen the lens on the metabolic damage.
