Objective1. To detect the location where CCR7 is expressed in colorectal cancer SW480 cells by immunohistochemisty.2. First, examine the changes of CCR7 expression in colorectal cancer SW480 cells, non-small-cell lung carcinoma SPC-A1 cells, and nasopharyngeal carcinoma CNE-2 cells after the cells were exposed to different X-ray irradiation doses after 24 and 48 hours; evaluate the CCR7 expression in SPC-A1 cells treated by a single 8Gy dose and 2Gy/day for 6 days that is equal to a single 8Gy in biology effect after 24 and 48 hours, respectively; investigate the effects of irradiation,chemicals and concurrent X–ray irradiation + chemicals on CCR7 expression in CNE-2 cells after 48 hours. Through these studies, observe the effects of irradiation, chemicals and concurrent X–ray irradiation+chemicals on the expression of chemokine receptor CCR7.Methods:1. Detect the location where CCR7 is expressed in SW480 cells by immunohistochemistry.2. Irradiate the SW480, SPC-A1 and CNE-2 cells with 0Gy, 2Gy, 10Gy, 20Gy X-ray doses respectively in single fraction, after 24 and 48 hours, examine the CCR7 expression.3. Irradiate the SPC-A1 cells with a single 8Gy and 2Gy/day for 6 days respectively, after 48 hours analyze CCR7 expression in these two groups.4. Observe the effects of Cisplatin(0μg/ml,0.032μg/ml,0.8μg/ml,20μg/ml)on the expression of chemokine receptor CCR7,after they are treated for 24 and 48 hours; compare the irradiation, chemicals and concurrent X–ray irradiation+chemicals on the expression of chemokine receptor CCR7, treat the CNE-2 cells with the control, 2Gy X-ray, 0.032μg/ml DDP, 0.8μg/ml DDP, 20μg/ml DDP, 2Gy+0.032μg/ml DDP, 2Gy+0.8μg/ml DDP and 2Gy+20μg/ml DDP, respectively. After 48 hours, the expression of CCR7 was tested.Results1. The expression of CCR7 is mainly located on cell membrane.2. In SW480 cells,the expression of CCR7 at 24 hours after irradiation increase significantly,at 48 hours after irradiation, the CCR7 expression in cells irradiated with 2Gy, 10Gy and 20Gy are much higher than that of the control, with no difference between different doses.3. In SPC-A1 cells the CCR7 experession dosen't change after irradiated with X-ray, regardless of the doses.4. The expression of CCR7 increases after treated by irradiation or Cisplatin. At the density of 0.8μg/ml DDP+2Gy, 20μg/ml DDP+2Gy, the concurrent chemoradiotherapy could reduce the CCR7 expression in CNE-2 cells compared with treated with Cisplatin alone.Conclusion1. CCR7 is mainly located on cell membrane in colorectal cancer SW480 cells.2. The CCR7 expression in SW480 and CNE-2 cells increases after exposed to X-ray. However, there are no effects of radiotherapy on the expression of CCR7 in SPC-A1 cells.3. Cisplatin could up regulate the CCR7 expression; However, after concurrent using X-ray irradiation and Cisplatin, the expression of CCR7 in CNE-2 cells reduced significantly than in Cispltin alone group.
