Objective: Ovarian cancer is a common malignant tumor of women reproductive system. About 90% is epithelial oarian cancer in ovarian tumors. Because of growth mode and location of ovarian cancer, it's difficult to diagnosis the early stage of ovarian cancer.Due to ovarian cancer developed easily and transfered widely,the 5-year survival rate is only 25% - 30%. Up to now,epithelial oarian cancer pathogenesis is still unclear.Perhaps it has relation to the change of certain gene expression level . KISS-1 gene is a tumor metastasis inhibitory factor. Some research show that the reduce or missing of KISS-1 Gene expression affect the tumor invasion and metastasis . Osteopontin ( OPN) ,to be a kind of phosphorylation glycoprotein, which contains saliva acid, is a kind of important extracellular matrix, some studies have found that it can promote the invasion and metastasis of tumors [1], but the exact mechanism is unclear. Recently, the other scholars found that OPN has an important role in the invasion and metastasis of some tumors, such as breast cancer, colon cancer, and pancreatic cancer, lung cancer ,prostate cancer and so on. However,Is there any relation OPN and KISS-1 in the invasion and metastasis of oarian cancer? So far,the report about the correlation of OPN and KISS-1 expressed in ovarian cancer has found little. This study aims to apply immunohistochemical stains method and explore the significance and correlation of KISS-1 and OPN expression in ovarian cancer cells .Methods:1 Immunohistochemistry was employed to measure the expression of KISS-1 and OPN in 43 patients with ovarian cancer tissue and 24 patients ovarian benign tumor tissue.2 Statistics:SPSS 13.0 was applied to analyze the results of experiment.P≤0.05 indicating significant difference.P≤0.01 indicating very significant difference. Results:1 Expression of KISS-1 and relation with clinical parameter Immunohistochemistry was employed to measure the expression of KISS-1 in 43 patients with ovarian cancer tissue and 24 patients ovarian benign tumor tissue.The positive expression rate of KISS-1 protein in ovarian cancer tissue was 39.53%(17/43), while in ovarian benign tumor tissue was 75.00%(18/24).There was very significant difference between the two groups(χ~2=7.765, P=0.005, P <0.01).The expression of KISS-1 gene protein in ovarian benign tumor tissue was significantly higher than that in the ovarian cancer tissue. The expression of KISS-1 protein in ovarian cancer tissue: The expression of KISS-1 gene protein in a lymph node metastasis group was significantly below Without lymph node metastasis group. (χ~2=4.036, P=0.045, P <0.05); The expression of KISS-1 gene protein in early cancer (Ⅰ,Ⅱstage ovarian cancer) was significantly higher than that in late-stage cancer (Ⅲ,Ⅳstage ovarian cancer) (χ~2=6.029, P=0.014, P <0.05);In different pathological type of ovarian cancer and different sicken age group, KISS-1 gene expression no obvious difference(P >0.05).2 Expression of OPN and relation with clinical parameterImmunohistochemistry was employed to measure the expression of OPN in 43 patients with ovarian cancer tissue and 24 patients ovarian benign tumor tissue.The positive expression rate of OPN protein in ovarian cancer tissue was74.42%(32/43) while in ovarian benign tumor tissue was 37.50%(11/24).There was very significant difference between the two groups(χ~2=8.841, P=0.003, P <0.01). The expression of OPN protein in the ovarian cancer tissue was significantly higher than that in ovarian benign tumor tissue. The expression of OPN protein in ovarian cancer tissue: The expression of OPN protein in a lymph node metastasis group was significantly higher than Without lymph node metastasis group. (χ~2=11.692, P=0.001, P <0.01); The expression of OPN protein in Late-stage cancer (Ⅲ,Ⅳstage ovarian cancer) was significantly higher than that in early cancer (Ⅰ,Ⅱstage ovarian cancer)(χ~2=6.029, P=0.014, P<0.05);In different pathological type of ovarian cancer and different sicken age group, OPN expression no obvious difference(P>0.05).3 Correlation of KISS-1 and OPN expression:In immunohistochemical protein expression level, there was negative correlation between KISS-1 and OPN expression(r=-0.507, P=0.001, P <0.01)Conclusions:1 The higher expression of KISS-1 in ovarian benign tumor tissue show that KISS-1 could inhibit the invasion and metastasis of ovarian cancer . The higher expression of OPN in ovarian cancer tissue show that OPN might be promote the invasion and metastasis of ovarian cancer of ovarian cancer.2 In ovarian cancer tissue,the expression of KISS-1 gene protein in a lymph node metastasis group was significantly below Without lymph node metastasis group; The expression of KISS-1 gene protein in early cancer (Ⅰ,Ⅱstage ovarian cancer) was significantly higher than that in late-stage cancer (Ⅲ,Ⅳstage ovarian cancer).It show that KISS-1genes could inhibit the invasion and metastasis of ovarian cancer.
