Background and objective:There are a lot of Studies on relationship of osteopontin with cancer cell in recent years,however studies on their related Integrin are still few. The present study intends to investigate the level of OPN,Integrinαvβ3 and VEGF expression and to evaluate the possible correlation between the expression of OPN,αvβ3 and VEGF and the clinic pathologic characteristics in the growth,invasiveness and metastasis of EOC,and seek possible tumor mark for EOC.Methods : Immunohistochemistry stain was used to detect the expression of OPN,αvβ3 and VEGF from 50 specimen obtained following operation of epithelial ovarian carcinoma patient,to analyze their interrelationship and its relation to the clinic pathologic factors.Results: OPN,αvβ3 and VEGF expression was mainly located in the cytoplasm of EOC cel1. Cytoplasm appeared yellow or brown which was identified as postive cell.The positive rates of OPN,αvβ3 and VEGF in 50 cases EOC tissues were 74.0%,64.0% and 72.0%,respectively.The positive expression of OPN,αvβ3 and VEGF were higher in every stage with the higher cinical stage;The positive expression of OPN,αvβ3 and VEGF protein showed significantly higher in patients with the advanced stages (clinical III~IV )compared to the early stages (clinical I~II ), There were significant difference in different cinical stage (P<0.01);The positive expression of OPN andαvβ3 protein showed significantly higher in patients with the poor and moderate histological grade than the good histological grade (P<0.01) ;The expression of VEGF did not appear significant difference in histological grade (P>0.05) ; There were not significant difference the positive expression of OPN,αvβ3 and VEGF in different histological type (P>0.05);The positive expression of OPN,αvβ3 and VEGF protein showed significantly higher in patients with lymde matasis than negative lymde matasis (P<0.01). The expression of OPN showed direct correlation toαvβ3 and VEGF expressions, and the expression ofαvβ3 had no correlation to VEGF expressions. The co-expression of OPN,αvβ3 and VEGF had remarkable correlation to with lymde matasis of EOC.Conclusion:1. The OPN,αvβ3 and VEGF protein was overexpressed in EOC tissue. The expression of OPN andαvβ3 had close relationship with the patients cinical period, histological grade and lymph nodes metastases of epithelial ovarian carcinoma; the expression of VEGF was remarkable related with clinical stage and lymph nodes metastases.It might reflects the progression of disease. 2.OPN may have an interaction withαvβ3 . It may play an important role in upgrading the expression ofαvβ3 in EOC .3.The expression of OPN showed remarkable correlation to VEGF expression,and it indicated that OPN maybe participate in carcinioma angiogenesis. 4.The expression of OPN showed direct correlation toαvβ3 and VEGF expressions ,they may act cooperatively in EOC. 5.To examine these 3 clinicalindicators in EOC is helpful to predict the progress and metastatic potential ,and to evaluate the prognosis for EOC,which indicates that OPN may be an early molecular event in the process of tumorigenesis and could be used as a biological marker to monitor the malignant process of EOC.
