| Objective To investigate the polymorphism of tumour necrosis factor -α(TNF-α)gene at Position一863,—857,—238 in Chinese Han population Anhui region ,and the association of which with Graves disease(GD) susceptibility.Then to explore the genetic back ground of GD.Methods466 cases were devided into two groups: Graves disease(GD) group,254cases(male 53,female 201)and control group,212cases (male 61,female 151).Genomic DNA were extracted from peripheral white blood cell. The polymorphism of tumour necrosis factor -α(TNF-α)gene at Position一863,—857,—238were determined by polymerase chain reaction and sequence specific primers ,via agarose gel electrophoresis to read allele and genotype. The frequencies distrubition of allele and genotype between GD group and normal control group,different sex group were compared by chi square,applying SPSS 16.0 statistical software.The frequencies distrubition of allele and genotype of different TRAb levels(TRAb﹥12U/L;≤12U/L) were also compared in earlier onset of GD.Results(l)The A allele frequeney (16.73%) of -863 locus in GD group was signifieantly higher than the frequeney(11.79%) in normal control group(P<0.05,OR=1.503);the frequeney of AA+CA genotype(32.68%) of -863 locus in GD group was significantly higher than the frequeney(23.58%) in normal control group(P<0.05,OR=1.573).There were no significant difference(p>0.05) of the frequencies distrubition of allele and genotype of -857,-238 locus between GD group and normal control group .(2) There were no significant difference(p>0.05) of the frequencies distrubition of allele and genotype of -863,-857,-238 locus between male GD group and female GD group .(3) There were no significant difference(p>0.05) of the frequencies distrubition of allele and genotype of -863,-857,-238 locus between different TRAb levels(TRAb﹥12U/L;≤12U/L) in earlier onset of GD.Conclusion(1)there are TNF-αgene—863C/A,—857C/T,—238G/A Polymorphisms in Chinese Han population Anhui region.(2) There is association of TNF-αgene -863 C/A polymorphism with GD susceptibility in Chinese Han population Anhui region.(3) There are no association between TNF-αgene—857C/T,—238G/A polymorphisms and GD susceptibility in Chinese Han population Anhui region.(4) There are no association between TNF-αgene—863C/A,—857C/T,—238G/A polymorphisms and male or female GD (5). There are no association between TNF-αgene—863C/A,—857C/T,—238G/A polymorphisms and TRAb level in earlier onset of GD. |
