Objective To research the protein and mRNA expression of a disintegrin and metalloproteinase 12 (ADAM12) in esophageal squamous cell carcinoma (ESCC), and to explore its role in carcinogenesis and development of ESCC .Methods One hundred cases of ESCC, 10 cases of pericancerous normal esophageal tissues, 18 cases of esophageal squamous cell dysplasia were detected for the expression of ADAM12 protein and mRNA by using immunohistochemistry (IHC) and in situ hybridization (ISH), respectively.Rusults1. The positive rates of ADAM12 protein in pericancerous normal esophageal tissues, esophageal squamous cell dysplasia and ESCC were 30.0% (3/10), 44.4% (8/18) and 82.0% (82/100). The positive rates of ADAM12 mRNA expression in pericancerous normal esophageal tissues, esophageal squamous cell dysplasia and ESCC were 40.0% (4/10), 50.0% (9/18) and 90.0% (90/100), ADAM12 protein and mRNA expression was significantly upregulated in ESCC group compared with that in normal esophageal tissues and in esophageal squamous cell dysplasia (P<0.05).2. No relationships were found between the expression of ADAM12 protein and mRNA and a series of clinical pathologial parameters of ESCC, in cluding histological grading, tumor size, infiltrating depth and lymph node metastasis (P>0.05).3. Positive corelation was found btween the expression of ADAM12 mRNA and its protein.Conclusion Protein and mRNA expression of ADAM12 is significantly upregulated in ESCC, and its biological significance remains to be further verified.
