Effect Of Ornithoctonus Huwena Venom On Proliferation Of Human Colon Carcinoma Cell SW480 In Vitro

Objective:To observe effect of Ornithoctonus huwena Venom on proliferation of human colon carcino cell SW480 in vitro to investigate antineoplastic activity of Ornithoctonus huwena Venom.Methods:human colon carcino SW480 cell line are cultured in vitro as the experimental model.Human colon carcino cell SW480 intervene by different concentration solution of Ornithoctonus huwena Venom in vitro,MTT chemosensitivity test evaluate suppression ratio and calculate IC50. Cell morphology Changes are directly observed and observed after dyeing by Hoechst 3334 through the microscope 2.Cells state and cells quantity are observed through flow cytometry after dyeing by AnnexinⅤ-FITC/PI.Cells changes of DNA are observed by agarosegel electrophoresis. spectrophotometry evaluate caspase-3 relative activating in cell.Results:MTT chemosensitivity test results show that human colon carcino SW480 cell quantity increase when intervene by lmg/ml concentration huwentoxin and SW480 cell quantity reduce when intervene by 5mg/ml～100mg/ml concentration huwentoxin, IC50=43.68mg/ml(48h).Microscopic observation results show that SW480 cell quantity reduce with part of the cell were broken and cells of various shapes.Dyeing by Hoechst 33342 results show that cell changes not only cell apoptosis but also cell necrosis, flow cytometry results also show that the proportion of dead cells increase with concentration increase and time prolong. Agarose gel electrophoresis results show that SW480 cell DNA degradation Spectrophotometry results show that activated caspase-3 in SW480 cell don't have distinct changes.Conclusion:1.effect of huwentoxin in sw480 cell both promote the proliferation and inhibits proliferation,but its main function is to inhibit proliferation; 2. effect of high concentration ornithoctonus huwentoxin in sw480 cell lead to both cell apoptosis and cell necrosis; 3. High concentration ornithoctonus huwena venom inhibit sw480 cell proliferation possible mechanism related to the degradation of DNA but not related to caspase-3 relative activation in cell...
