Objective:Lung cancer is one of the most common malignant tumors, and the tumor metastasis is the main reason to cause patients died. Researching the transfer mechanism can provide effective strategy for lung cancer prevention and treatment. IL - 8 is a chemotactic factor, also a promote angiogenic factor. It can promote many kinds of tumor metastasis, for example melanoma and stomach cancer. But the mechanism is not clear in lung cancer. This article is to investigate the effect and mechanism of IL-8 on the cell migration in A549 cells (Human lung adenocarcinoma epithelial cell line).Method:(1)The effect of IL-8 on cell viability was measured by MTT arrays. (2)The effect of IL-8 on cell migration was measured by Wound-healing experiment and Transwell arrays, respectively. (3)The effect of IL-8 on the expression of MMP-2,MMP-9 and p-JNK/SAPK, and on the expression of MMP-2 by influence of C-JNK/SAPK inhibitor(SP600125)was detected by Western blot assays, respectively. (4) The effect of lower expression of MMP-2 on A549 cells migration was detected by Wound-healing experiment.Results:The appropriate concentration of IL-8 in experiment was chosen by MTT arrays. Wound-healing experiment and Transwell arrays were used to prove that IL-8 can promote the migration of A549 cells. The results of Western blot showed that:(1) IL-8 can promote the expression of MMP-2 protein, but no significant effect on MMP-9 expression.(2) IL-8 can promote the expression of p-JNK/SAPK.(3) Inhibitor(SP600125)can block the impacts of IL - 8 to MMP-2 expression. Wound-healing experiment proves that lower expression of MMP–2 can inhibit A549 cells migration.Conclusion:IL-8 can regulate the expression of MMP-2 protein and further promote the migration of A549 cells through the JNK/SAPK signaling pathway.
