| Objective:To investigate the relation between the anti-depression effect of reboxetinel and the expressions of CORT,BDNF,NET,and 5-HTT in rat model-induced by CUMS.Method:1 60 male SD rats were randomly divided into following groups:normal group(NG),model group(MG),normal group-treated with 0.7 mg/kg/d reboxetine(RNG),and model group-treated with reboxetine(RMG). Chronic mild unpredicted stress(CUMS) with solitary condition was taken to establish rat depression model.2 The open-field test,sucrose consumption,and elevated plus maze(EPM)assay were used to evaluate the depression behaviors of rats. Biochemistry methods were used to determine the content of MDA and activities of SOD and CAT in both of serum and brain cortex. Radioimmunity method was used for determination of the corticosterone (CORT) concentration in rat serum. Immunohistochemistry staining was taken to determine BDNF protein expression of hippocampus. Reverse transcription polymerase chain reaction (RT-PCR) method was used to determine the mRNA expressions of NET, 5-HTT,BDNF, CRF in pons cerebelli, hippocampus, and hypothalamus tissues of rats. Western-blot method was taken for determining the protein expressions of NET and 5-HTT in pons cerebelli and hippocampus tissues of rat.3 Hippocampal pathological change with HE staining was observed under microscope.Results:1 It was shown that there were significant decreases of crossing,rearing,and preening activities from open-field test,sucrose consumption,and activity and retention time from elevated plus maze in model group in comparison of NG. The treatment of roboxetine obviously improved the symptoms caused by CUMS. However treatment of roboxetine had no influences on behaviors of NG rats.2 CUMS significantly increased MDA content and reduced SOD and CAT activities of serum and cortex,elevated serum CORT concentration,decreased expressions of NET,5-HTT,and BDNF,and promoted CRF expression,from the variety of brain areas. The treatment of roboxetine obviously blunted the changes above. However treatment of roboxetine had no influences on MDA content,SOD and CAT activities,CORT concentration,and expressions of NET,5-HTT,BDNF,and CRF of NG rats.3 An obvious karyopyknosis of hippocampal neurocytes was observed in model group,compared with NG. The treatment of roboxetine significantly relieved the pathological changes.Conclusion:The treatment of reboxetine can improve depression behaviors of rats induced by CUMS through reversing imbalance of oxidative stress and anti-oxidative stress systems,decreasing CORT and CRF level,and increasing expressions of NET,5-HTT,and BDNF. |
