| Objective: To observe the changes of BDNFã€IDOã€CORT and Treg levels in the peripheral blood among patients with depression and health control, the interactional and correlational analyses were conducted,to found related pathway which could result in depression,to provide new theoretical basis for pathogenesis of depression.Methods: From September 2013 to January 2014, 60 cases of the hospitalized in Guiyang First People,s Hospital and Affiliated Hospital of Guiyang Medical College were chosen as the depression group.The normal control group included 30 healthy volunteers from Affiliated Hospital of the Guiyang Medical College. Flow cytometry was used to detect Treg. We tested the samples of both the depression group and the healthy control group via ELISA in accordance with the instruction and measured OD on microplate reader and calculated the serum concentration of BDNFã€IDO and CORT.Statistical software SPSS 11.5 was used to process statistical analysis for the test value of aforementioned Index. Results: a comparison of controls and patients with depression,results as follows: 1.The BDNF concentration detected by serum of depression group is(6.72±1.33)μg/L,the concentration is considerably below the level of(13.11±2.16)μg/L in healthy control group,P=0.000. The IDO concentration detected by serum of depression group is(83.43±12.03)μg/L, the concentration is higher than the level of(49.41±11.70)μg/L in healthy control group,P=0.009.The CORT concentration detected by serum of depression group is(185.43 ±32.94)μg/L, the concentration exceed the level of(124.45±14.38)μg/L in healthy control group,P=0.000.The Treg cell,s percentage of depression group in PBMC is(7.6±1.5)%,the ratio is higher than(4.7±0.9)% in healthy control group,P=0.004. All of these difference between depression group and control group was statistically significant(P<0.01). 2.All of these depression indexes were on differences in difference patient’s condition and course of disease,these indexes include the concentration of BDNFã€IDOã€CORT in serum and the ratio of Treg in peripheral blood. 3.The respective correlation of depression group and control group is as follows. in the control group: BDNF and IDO(r=0.433,P=0.017),BDNF and CORT(r=0.547,P=0.002),BDNF and CORT(r=0.454,P= 0.012).In the depression group: BDNF and IDO(r=-0.825,P=<0.001),BDNF and CORT(r=-0.688,P<0.001),BDNF and Treg cell(r=-0.667,P<0.001),IDO and CORT(r=0.804,P<0.001),IDO and Treg cell(r=0.753,P<0.001),CORT and Treg cell(r=0.812,P<0.001). Conclusions:IDO plays an important role in the pathogenesis of depression, elevated IDO closely relates the increased CORT levels resulted in stress,the decreased BDNF levels and the increased Treg ratio together,it is an important part of pathogenesis in depression. |
