Object To investigate the expression of transient receptor potential canonical 3(TRPC3) and transient receptor potential canonical 6(TRPC6) in the temporal cortex from patients with intractable temporal lobe epilepsy(TLE) and explore their potential roles in epileptogenesis.Methods This study was based on 20 patients with intractable temporal lobe epilepsy and 6 controls with acute head trauma.Temporal cortex tissue was obtained from each subject. Immunohistochemistry and Western blot were performed to examine the expression of TRPC3 and TRPC6 in each group.Results The immunohistochemisty study showed that TRPC3 and TRPC6 were mainly located in the cytomembrane and cytoplasm of neurons. Immunohistochemistry and Western blot analyses showed increased expression of TRPC3 and TRPC6 in patients with intractable TLE compared with controls.Conclusions The expression of TRPC3 and TRPC6 was upregulated in the temporal cortex tissue of patients with intractable TLE, which suggests their potential roles in epileptogenesis.
