Objective: Fascin-1is a cytoskeleton protein that is widelyexpressed in the central nervous system (CNS). It plays important roles incell morphology and function. Many studies have found that Fascin-1istightly related to synaptic plasticity, and abnormal Fascin-1regulation isassociated with many CNS diseases. Synaptic plasticity is an importantpathological foundation for temporal lobe epilepsy (TLE), however, it isstill not clear whether Fascin-1is involved in the pathogenesis of TLE.Hence, the aim of this study was to investigate Fascin-1expression inpatients with TLE and in a rat model of pilocarpine-induced epilepsy.Methods: We randomly selected20cases of TLE foci brain tissuesamples from the brain tissue library and10temporal cortex samples withnormal tissue structure were used as controls in our study.40male SD ratswere randomly divided into control group(5) and experimental group(35).Rats were injected intraperitoneally with pilocarpine to establish TLE rat model. All rats were sacrificed at different time point (6h,24h,72h,1W,2W,1M,2M)after kindling. Immunochemistry, western blot andimmnofluorescence methods were used to assess the expression andlocation of fascin-1in cortex and hippocampus.Results: Fascin-1is mainly expressed in the cytoplasm and cellmembrane of cortex of the temporal lobe and hippocampal neurons.Fascin-1expression in patients with TLE was significantly lower than thatin the normal controls. Equally, Fascin-1expression in the temporal lobecortex and hippocampus of the rat model was lower than that in the controlgroup at all time points (but not6h time-point of cortex). The lowestFascin-1expression in the cortex was observed at2months, and the lowestexpression in hippocampus was observed at2weeks.Conclusions: Fascin-1protein expression in patients with TLE and inthe pilocarpine-induced epilepsy rat model was down-regulated, suggestingthat Fascin-1may be involved in the pathogenesis of TLE.
