Objective:The purpose of this study is to detect the expression of VEGF in KS of head and neck, and analyze HHV-8 infection how to affect the VEGF expression by using molecular biology and cell biology methods, so as to investigate its role in the pathogenesis of KS of head and neck. Methods:We Collected tumour tissues and vicinal normal tissues from 13 cases of KS patients of head and neck in xinjiang, including 3 cases of Frozen tissues,6 cases of Paraffin-embedded tissues and 4 cases of paraffin section, then applied Reverse transcription-polymerase chain reaction (RT-PCR) technique to detect VEGF mRNA expression in 3 cases with KS of head and neck and 3 cases with tumor-free margins as control. S-P immunohistochemical method was used to detected the VEGF,CD34 and LANA protein expression in 13 cases with KS of head and neck and 9 cases with tumor-free margins as control. Western blotting was used to detected the expression of VEGF in BCBL-1(HHV-8+) and BJAB (HHV-8-) cells. Results:The higher mRNA and protein expression level of VEGF were observed in KS tissues of head and neck than in vicinal normal tissues (P<0.01); The higher expression of CD34 were observed in KS than in normal tissue(P<0.01); there were positive correlations between VEGF expression and microvessel density(MVD). HHV-8 infection was detected in all KS tissues of head and neck, and the expression of VEGF in BCBL-1 cells was higher than that in BJAB cells(P<0.01). Conclusions:VEGF is overexpressed in KS of head and neck, VEGF and CD34 are significant correlated with angiogenesis and development of KS. HHV-8 infection exists in all KS tissues of head and neck, and VEGF overexpression is closely correlated with HHV-8 infection. Inhibiting VEGF expression in KS tissues and combining anti-herpesvirus therapy maybe provide a new path and evidence for KS prevention and cure.
