Objective:To explore the correlation between CD4+ CD25+ Foxp3+ regulatory T cells and TGF-(3,.1L-10 through testing the change of CD4+ CD25+ Foxp3+ regulatory T cells,TGF-β1 and IL-10 in peripheral blood of esophageal cancer patients in Hazak according to different TNM stages. Methods:The study was carried out in a total of 47 Hazak patients with esophageal cancer diagnosed by Xinjiang Medical University affiliated Tumor Hospital from January to November in 2010.All patients were newly diagnosed and none had received any form of anti-cancar therapy before collection of their peripheral blood for test.30 healthy Hazak volunteer were included in this study as the control group.1. Flow cytometry was used to analyze the proportion of CD4+ CD25+ Foxp3+ regulatory T cells in CD4+ T cells in peripheral blood mononuclear cells (PBMCs) of 47 Hazak patients with esophageal cancer and 30 healthy peoples.2. enzyme-linked immunosorbent Assay (ELISA) was used to detect the level of TGF-β1,IL-10 in 47 Hazak patients with esophageal cancer and 30 healthy peoples. Results:1. the proportion of Treg cells, the expression level of TGF-β1 and IL-10 in the case group were obviously higher than that of the control group, the difference has statistics significance (P< 0.05).2. compared with control, there were not statistically significant difference of the proportion of Treg cells, the expression level of TGF-β1 and IL-10 in phase of TNM stage (Ⅰ~Ⅱ) and without lymph node metastasis (P>0.05), there were statistically significance of the proportion of Treg cells, the expression level of TGF-β1 and IL-10 in phase of TNM stage (Ⅲ~Ⅳ) and lymph node metastasis (P<0.05); the difference of the proportion of Treg cells, the expression level of TGF-β1 and IL-10 between the stage of TNM(Ⅰ~Ⅱ) and (Ⅲ~Ⅳ) was significant (P<0.05).3.the expression level of TGF-β1,IL-10 was significantly correlated with the proportion of CD4+ CD25+ Foxp3+ regulatory T cells (P<0.01).Conclusion:1. the proportion of Treg cells in peripheral blood of the Hazak esophageal patients significantly increased, it probably had certain value in judging disease stage of esophageal cancer in Hazak by detecting the proportion of Treg.2. the expression level of TGF-β1,IL-10 were obviously increased,it was probably that TGF-β1 and IL-10 could inhibit the body's anti-tumour immune responses.3. the expression level of TGF-β1, IL-10 were significantly related to Treg cells, and it may be involved in the immune negative adjustment mechanism of body's.
