Effects And Mechanism Of Hypertonic Saline On Rat's Liver Warm Ischemia-reperfusion Injury

ObjectiveThe purpose of this paper was to study the effects of Hypertonic Saline on rat 's liver warm ischemia reperfusion injury (HIRI) and compare them with ischemia preconditioning to explore the mechanism.MethodsEighty clean and healthy male rats were randomly divided into four groups sham operation group,ischemia reperfusion group, HTS group and ischemia preconditioning group ,twenty respectively. HIRI rat models were established by Pringle′s method in all group to determine the levels of serum AST(alanine-aminotransferase),ALT(alnine-aminotransferase),LDH(lactate-dehydrogenase),SOD(superoxide-ismutase),Bax and Bcl-2's expression level at the four time point 1,3,6 and 24 hour of reperfusion,and to observe the changes of liver histopathology and ultrastructure,and calculate AI(Apoplosis Index) to observe the hepatocellular apoptosis by TdT-mediated dUTP-biotin nick end labeling (TUNEL).ResultsAt 1,3,6,24h after reperfusion,the serum levels of ALT,AST,LDH ,and the expression of Bax and AI of group IR , group HTS and group IP were significantly higher than that of group Sham , in those group of HTS and IP were lower than group IR, group HTS is the lowest (P<0.01),however, SOD levels and Bcl-2 expression level of group IR, group HTS and group IP were significantly lower than group Sham , Those of group HTS and group IP were higher than group IR, Those of group HTS was the highest (P<0.01). In group HTS and group IP , the pathological and ultra-structure changes of liver were significantly sligher than group IR. The group HTS is the slighest.ConclusionsHypertonic saline showed a preventive effect on hepatic ischemia reperfusion injury just as IP , by increaseing the expression level of Bcl-2 and decreaseing that of Bax and hepatocellular apoptosis. Hypertonic saline may have a better effect than ischemia preconditioning .
