PurposeTo explore the osteoporosis treatment function and the mechanism of treating osteoporosis by detecting the proteins and gene expression of OPG and RANKL.Material and MethodsThe experiment is randomly divided into 5 groups: normal control group (12 rats), sham-operated group (12 rats), model group (12 rats), positive control group (13 rats), and exercise group (11 rats). Remove the ovaries of rats of Model group, positive control group and exercise group. Do the same operation on rats of Sham-operated group; the only difference is that we only remove small adipose tissue not the ovaries.A month later after operation, the rats of exercise group start doing moderate intensity exercise. Feed the positive control group rats with diethylstilbestrol, feed the normal control group, sham-operated group, model group and exercise group rats with distilled water, do exercise for five days a week, rest for two days, again for five days, continue for 3 months.In order to mark the bone with fluorescence, at the third and the sixteenth day before the day which all the rats were killed, hydrochloric acid tetracycline were injected into the abdomen of the rats according to 30mg/kg. The rats were stripped the tibia after being guillotined. Fix the tibia with neutral polyphosphate formaldehyde ,the left tibia were retained for Demineralization bone frozen section, while the right tibia were retained for bone tissue section which were not decalcified, which were made for index detection.ResultsAfter ovariectomy, compared with normal control group rats, TBV% of model group rats were significantly decreased, after taking exercise, TBV% significantly raised. TRS%, TFS%, AFS%, MAR, mAR and OSW of normal control group rats were significantly higher than model group rats, while significantly lower after taking exercise.After ovariectomy, OPG protein and mRNA expression of OB and MSC were significantly decreased, RANKL protein and mRNA expression were significantly raised. After taking exercise, OPG protein and mRNA expression of OB and MSC were significantly raised, RANKL protein and mRNA expression were significantly decreased.Conclusion:1 Taking exercise has a certain improving effect on indicators related to histomorphometry.2 Taking exercise plays a regulatory role in proteins and gene expression of OPG and RANKL.3 Taking exercise has a certain preventive effect on osteoporosis caused by ovariectomy.
