Objective: This study was designed to evaluate the expression of HBD-2 and mPGES-1 in human normal salivary gland, salivary gland tumor and inflammation salivary gland tissuses, and determine their relationships and correlations with clinicopathologic parameters of salivary gland cancer, and to study the regulation of HBD-2 expression by PGE2.Methods: The expression of HBD-2 in salivary gland benign tumor, salivary gland cancer, inflammation salivary gland tissuses and normal salivary gland tissuses were detected by RT-qPCR and immunohistochemical staining. Then their relationships and correlations with clinicopathologic parameters of salivary gland cancer were analyzed. The expression of HBD-2 mRNA was evaluated by RT-qPCR in ACC-M cells treated by different concentration of PGE2.Results: RT-qPCR results showed that, compared with normal tissues, the HBD-2 mRNA expression in salivary gland benign tumor, salivary gland cancer and inflammation tissues were amount to 6.468 fold, 0.334 fold and 10.563 fold respectively (p<0.05). The mPGES-1 mRNA expression in salivary gland benign tumor, salivary gland cancer and inflammation tissues were amount to 0.896 fold (p>0.05), 8.326 fold and 11.581 fold respectively (p<0.05). The HBD-2 expression in salivary gland cancer was correlated with several lymph node metastasis and TNM stage (p<0.05), but not related to histology primary tumor size, patients'age and other clinicopathologic parameters (p>0.05). Compared with control group, the HBD-2 mRNA expression in group 10nmol/ml, group 50nmol/ml, group 100nmol/ml, group 200nmol/ml and group 1μmol/ml were amount to 1.142 fold, 2.364 fold, 11.917 fold, 13.114 fold and 0.453 fold respectively (p<0.05).Conclusion: In salivary gland benign tumor tissues and inflammation tissues, HBD-2 mRNA and protein expression were significantly increased than in normal salivary gland tissues; in salivary gland cancer tissues HBD-2 mRNA and protein expression were significantly decreade than in normal salivary gland tissues and significantly associated with clinicopathologic parameters. In salivary gland cancer tissues and inflammation tissues, mPGES-1 mRNA expression were significantly increased. HBD-2 mRNA expression was varible according to the consentration of PGE2, which implies some connection between the expression of HBD-2 and mPGES-1.
