Purpose: Preliminary analysis of the composition of steroid compounds of Oviductus Ranae.Isolate,purify and identify steroid compounds from the Oviductus Ranae. Study the synthesis of the compounds and determine the optimal method.Establish the content of these compounds by HPLC method of Oviductus Ranae.Method: In this paper, gas chromatography - mass spectrometry is used to analyze the steroidal compounds of the Oviductus Ranae medicine. By using CO2 supercritical extraction technology,we extract oil Constituents from the Oviductus Ranae. Purify and separate the dregs through SephadexLH-20, C-18 RP silica gel, HPLC and other modern separation, identify by spectroscopy, through the biological transformation and synthesize the steroidal compounds and using HPLC to study the determination of Oviductus Ranae. Result: By using gas chromatography - mass spectrometry we analyze preliminary 8-steroidal compounds: they are Cholest-5-en-3-ol,Cholestan-3-one,Cholesta-3,5-dien-7-one,Campesterol,Cholesta-4-dien-3-one,Cholesta-4,6-dien-3-one,Stigmasterol-22,23-dihydro-,Cholesta-8-dien-7-one.The steroids HI separated and purified by the SephadexLH-20, C-18 RP silica and HPLC and by using 13C-NMR and 1H-NMR spectroscopy and other modern test technology combined with ESI-MS Mass Spectrometry identify the structure of the Steroidal compounds is 4-Cholestene-3-one. Synthesized by biological transformation ,the conversion rate of the Steroidal compounds reached 90.2%.Measure the content of the compound of oviductus Ranae from different places,we get the result as following:Jingyu(7.24ug/g),Huadian(7.60 ug/g),Shulan(7.96ug/g).Conclusion: The compounds,which were got by GC-MS is of further guidance in the follow-up study of steroidal compounds of Oviductus Ranae. In this study, we extract and isolate the steroidal compounds and prepared it by using the biological transformation, which provide the material basis for its applied research. The establishment of the determination method of 4-Cholestene-3-one of oviductus Ranae has improved the quality control of medicines. The results of this study provides a basis for follow-up research, development and utilization of Oviductus Ranae.
