Objective:To develop a young male rat model of diet-induced obese and type II diabetic by high fat diets, and to study the effect of aerobic exercise and drugs (metformin) on protein expression of leptin and leptin receptors in skeletal muscle,this study tried to explore the mechanism that exercise and metformin improves leptin resistance.Methods:1.77 male rats just weaned were randomly divided into 7 groups: normal control group, diabetes control group,campaign treatment group,prevention campaign group,prevention treatment and campaign group, drug treatment group, drug and campaign treatment group. 11 rats each group.The normal control group rats were feeded normal diet for 12 weeks. The groups except normal control group were feeded high glucose and high fat diet for 12weeks.2.Diabetic rats in prevention campaign group,prevention treatment and campaign group were swimming trained six weeks.Diabetic rat model were induced by intraperitoneal injection with strep to zotocin (STZ) at the end of the sixth week,then,diabetic rats in diabetes control group,treatment campaign group drug treatment group,campaign and drug treatment group were swimming trained six weeks.3.After 12 week-interventions,all the rats were killed and leptin and leptin receptors in skeletal muscle were determined.Results:1.Compared with normal control group,diabetes control group had significant increased weight(P<0.05), blood sugar(P<0.01), insulin(P<0.01). Compared with normal control group, diabetes control group had increased TG(P<0.01) , FFA(P<0.05) in skeletal muscle. Leptin(P<0.01) in skeletal had significant increased,leptin receptors(P<0.05) in skeletal muscle had significant decreased .2.Compared with diabetes control group,prevention treatment and campaign group, drug campaign group had significant decreased fasting blood glucose ( P<0.01P<0.01)and Insulin(P<0.01) (P<0.01). Compared with diabetes control group,prevention treatment and campaign group, drug campaign group had decreased TG(P<0.01) (P<0.05), FFA(P<0.01) (P<0.01) in skeletal muscle. Leptin(P<0.05) (P<0.01)in skeletal muscle had significant decreased, leptin receptors(P<0.01) (P<0.05)in skeletal muscle had increased .3.Compared with campaign treatment group,drug and campaign treatment group had skeletal muscle decreased fasting blood glucose(P<0.05) and TG(P<0.01), FFA(P<0.05) in skeletal muscle.Leptin(P<0.05) in skeletal muscle had skeletal decreased,leptin receptors(P<0.05) in skeletal muscle had significant increased .Conclusion:1.High-fat diet with low-dose STZ-induced type 2 diabetic rat skeletal muscle leptin levels rise, reduced the protein expression of its receptor, triggering glucose and lipid metabolism disorder, there leptin resistance and insulin resistance.2.The long-term aerobic exercise training and drug intervention can be different degrees of skeletal muscle by reducing leptin levels, increases expression of its receptor protein content,to improve glucose and lipid metabolism disorders, alleviate the leptin resistance and insulin resistance, serve as an effective type 2 diabetes prevention, treatment and prevention effects, including prevention exercise group and exercis the role of combined medication group better.
