Honeysuckle is Caprifoliaceae Lonicera japonica Thunb dry bud,which is a traditional Chinese medicine,with a variety of biological activities.The main functional component of chlorogenic acid has antibacterial,anti-virus, anti-tumor,nourish the liver and gallbladder,reduce blood pressure,lipids,clearing free radicals and excited the central nervous system,is one of the active ingredient in many Chinese herbal medicines,but also certain medicine qualitative index.Chlorogenic acid is a very broad range of applications,mainly in medicine,daily chemical and food industries.This paper mainly studies chlorogenic acid detection methods,extraction and purification methods,and anti-oxidation,to provide a reference for improving the extraction purity and developping its functions.In this paper,the material based on honeysuckle of the Jinfeng 1st from Henan Fengqiu is comparatively studied by the spectrophotometer,high performance liquid chromatography,thin layer chromatography method of these three common chlorogenic acid detection methods.Conclusion:The HPLC because of its high-precision, accuracy,with spectrophotometry used for quantitative analysis,while TLC is more suitable for rapid qualitative analysis.In this study,the recent years of chlorogenic acid in Lonicera japonica common extraction process:aqueous extraction,alcohol extraction,ultrasonic extraction,recipitation method,etc.were compared,using high performance liquid chromatography method as the detection means,obtained improving extraction method - improved ethyl acetate method,the extraction rate was 37.7%,yield of 44.3%.Chlorogenic acid were isolated from Honeysuckle extraction at a time by high-speed countercurrent chromatography.The two-phase solvent system was chosen out:ethyl acetate:acetic acid:water(4:1:5, V/V),and optimizd the separation parameters:the flow rate is 1ml/min,instrument rotational speed is 800rpm,the temperature is 45℃.The method is simple and high separation efficiency,and can be used as a high-purity chlorogenic acid separation and preparation method.The antioxidation effect of honeysuckle extraction and the chlorogenic acid was studied.Comparing with Vc,the antioxidation activity was studied by reducing abilities,hydroxyl radical system and DPPH system.All the results above showed that the major antioxidation of honeysuckle extraction was chlorogenic acid;Honeysuckle extraction had strong capacity to scavenge HO·and DPPH·and chlorogenic acid,But the reducing capacity of chlorogenic acid was superior than VC and honeysuckle extraction.
