Objective: To study the effects of Extract from Rabbit Skim Inflamed by Vaccinia Virus for Injection (ERSIVVI) in fracture healing, and to explore its mechanisms in promoting the fracture healing . Methods: On the one hand, fracture models were established by operation at the middle shafts of both radii of experimental rabbits leaving a complete 3 mm bone defect in each side , the rabbits were randomly divided into three groups: the saline blank group,the experimental group of ERSIVVI and the positive control group of Cervus and Cucumis Polypeptide Injection(CCPI) , then each group was divided into two sub-groups which the biopsys were colleted on the 16 day and 29 day , each group was injected with equivalent saline,ERSIVVI and CCPI in turn at the bone defect. Examination items included: the general condition of fracture,serum biochemistry,X–ray score,bone mineral density(BMD),figure of callus,Biomechanics and Histomorphomety; On the other hand, the tibial bone fracture models were established by operation in SD rats , then the rats were randomly divided into five groups: the saline blank group,the experimental group of ERSIVVI with three differ level : 0.24U/ml,1.2U/ml and 2.4U/ml,and the positive control group of (CCPI) , then each group was divided into six sub-groups which the biopsys were colleted on the 1,2,3,4,5,6 week(1,2,3,4,5,6W) , each group was injected with equivalent saline,three differ leve(0.24U/ml,1.2U/ml and 2.4U/ml )ERSIVVI and CCPI in turn at the bone defect. the methods of histology and immunohistochemistry were used to detect the expression level of Nerve growth factor ( NGF ),Bone morphogenetic protein(BMP-2),Transforming growth factor-β1(TGF-β1),Vascular endothelia growth factor(VEGF)and CollagenⅠ(COL-Ⅰ)in the callus tissue during different period of fracture healing , then we compared and analyzed the data by measurement . Results: the experimental group of ERSIVVI had significant differences with the saline blank group, the experimental group of ERSIVVI was roughly the same with the positive control group of CCPI except NGF . Conclusion:ERSIVVI could remarkably promote the fracture healing , its effect was related with the dose, and its role was similar with CCPI .
