Objective:To investigate the significance of the loss of heterozygosity(LOH)of APC gene , the member of Wnt singnaling pathways,in adrenocortical adenoma.Methods:42 adrenocortical adenomas and 4 colon carcinoma was collected by us when these patients were suffering operation in our hospital.And the peripheral blood of each patient was collected before the operation. All tumor samples were confirmed by pathology .After that, we extracted the genome DNA of each tumor sample and each peripheral blood sample. Loss of heterozygosity (LOH) of adenomatous polyposis coli (APC),a component of the Wnt signaling pathways,was investigated by using Polymerase Chain Reaction/ Restriction Fragment Length Polymorphism(PCR-RFLP) in 42 adrenocortical adenomas (experimental group) and 4 colon carcinoma (positive control group) by two genetic markers,Rsa I in the exon11 and Msp I in the exon15 of APC. Results:A 133bp fragment was amplificated from the 11 exon and a 550bp fragment was amplificated from the 15 exon of the APC gene by Polymerase Chain Reaction. In 42 adrenocortical adenoma samples(experimental group) , 30 were informative (30/42,71.4%),but none of them show loss of heterozygosity (LOH).In 4 colon carcinoma (positive control group), 2 were informative and both of the two samples show loss of heterozygosity(LOH).Conclusions: None of the adrenocortical adenomas show loss of heterozygosity (LOH),it is supposed that the loss of heterozygosity (LOH) of APC gene might not be a common phenomenon in the development of adrenocortical adenomas.
