Ginsenosides is one of major ingredients of Panax ginseng.Many studies demonstrated that ginsenosides played important roles in anti-tumor, anti-oxidation, anti-fatigue, anti-apolexis and enhance organism immunity. Latest studies showed that panax ginseng used as Chinese herbal medicine immunopotentiators more often than ever, However, almost studies of ginsenosides stand in the situation of total saponin of Panax Ginseng or monomer ginsenoside, but the different ginsenosides of It's constituent were reported little by consulting literature. In order to obtain the different ginsenosides of It's constituent, panaxginseng was extracted with methanol solution, then mixed ginsenosides and macroporous adsorption resinto filled separating column, eluted with distilled water and concentration of 10%, 30%, 50%, 70% ethanol. High performance liquid chromatography in rapid determination of five ginseng extracts in the root of Panax Ginseng. The detection results show that the total amount of ginsenosides was maximum in the ginseng extracts eluted with 50% ethanol, but the ginseng extracts eluted with 70% ethanol has ginsenoside Rg3, is distinguished from other groups.The splenic lymphocyte transfer test was used to detect the lymphocyte proliferation effects of ginseng extracts and It's constituent in vitro.Splenic lymphocyte was collected and stimulated by ConA and LPS. Proliferative response of splenic lymphocyte was examined by MTT method. ginseng extracts and It's constituent separately or being combined with ConA and LPS showed to stimulate the proliferation of lymphocytes at the experimental concentration. Compared to the control group, Cell proliferation ability in ginseng extracts and eluted with 70% ethanol group showed the most significant effect as compared to the control (p <0.01).To investigate the effect of ginseng extracts and it's constituent on peritoneal macrophage in mice, The function of macrophage englobing neutral red was determined by neutral red trial. Ginseng extracts and it's constituent separately or being combined with LPS showed to stimulate the activity of macrophage englobing neutral at the experimental concentration. Compared to the control group. Peritoneal macrophage englobing neutral ability in ginseng extracts and eluted with 70% ethanol group showed the most significant effect as compared to the control(p <0.01).Sixty mices with 4~5weeks, body weight18~21g were randomly divided into 6 groups. They were abdominal cavity injected in with 0.2 mL mink enteritis parvovirus inactivated vaccine, mink enteritis parvovirus inactivated vaccine mixed with 125μg, 250μg, 500μg ginseng extracts eluted with 70% ethanol. A booster injection was administered 2 weeks after the Primary inununization. Blood samples were collected 3 days before the primary injection and 4 weeks after the booster injection. HI test was used to evaluate anti-Mink Parvovirus Enteriti antibody titers in the sera. The results showed that ginseng extracts eluted with 70% ethanol and aluminum hydroxide could induce higher anti-Mink Parvovirus Enteriti antibody in titers in sera. Aluminum hydroxide had stronger adjuvant effect than ginseng extracts eluted with 70% ethanol, but the mixture of ginseng extracts and aluminum hydroxide had strongest effect. Moreover sixty mices injected with ginseng extracts showed no side-effect at the injection sites. Thus, ginseng extracts was safe to use in mices. So ginseng extracts had significant adjuvant effects on humoral and cell-mediated response and were safe after the injection. The activity of mixture between ginseng extracts and aluminum hydroxide is better than that use only of ginseng extracts or aluminum hydroxide.
