Experiment Study Of Prescription Of ShuGanAnShen In Treatment Of Insomnia

Objective:To study the effect of prescription of ShuGanAnShen for the rats of sedative and hypnosis.Materials and Methods:120 Kun Ming healthy rats were evenly divided into three groups randomly. 40 rats in the first group were evenly divided into four groups randomly: control group, estazolam group, large dose of ShuGanAnShen group and small dose of ShuGanAnShen group. Rats in four groups were perfused by the same volume of distilled water, estazolam solution, large dose of ShuGanAnShen decoction, small dose of ShuGanAnShen decoction respectively. After seven days, each group was observed respectively to learn the prescription effect in aspect of autonomic activities. After this procedure, blood was taken from the rats in large dose of ShuGanAnShen group for biochemical assay. Then the rats were killed in order to get liver biopsy. 40 rats in the second group were grouped and irrigated medicine as the first group. Seven days later, the rats were perfused sodium pentobarbital above the threshold by celiac cavity and observed the effect of ShuGanAnShen in sleep latent time, sleep duration. The third group was as the same as the second group except that sodium pentobarbital were given subthreshold.Results:The prescription of ShuGanAnShen and Estazolam both could reduce the frequency of rats'autonomic activities,there was no significant difference between large and low dose group of ShuGanAnShen. The two drugs could also apparently decrease sleeping incubation period and prolong sleep duration. Estazolam and large dose group of ShuGanAnShen could increase the rate of falling asleep and there was no significant difference between the two groups. No effect of biochemical indexes and toxicity of liver of rats were discovered during the treatment by prescription of ShuGanAnShen.Conclusions:Prescription of ShuGanAnShen could decrease the frequency of rats' autonomic activities and period of falling asleep. It could also prolong sleeping time and increase the rate of falling asleep of rats. The prescription had an obviously effect on sleeping and no effect on liver and biochemical indexes .This prescription had effective treatments on insomnia.
