Objective: To investigate the effect of quercetin on the invasion and metastasis abilities of human colon cancer cell SW480 in vitro;Methods: Human colon carcinoma cell line SW480 were incubated in vitro, using cell biology techniques. The morphological changes of SW480 cell were investigated through light microscope and electron microscope; the effect of quercetin on SW480 cell was studied by means of CCK-8; Flowcytometry was used to study the pro-apoptosis and the cell cycle effects of quercetin on SW480 cells; and the adhesion and invasion of cells was tested using adhesion experiments and Transwell method; Gelatin zymography was also performed in quercetin treated SW480 cells.Results: The amount of human colon carcinoma cells treated by quercetin decreased, compared with that of controls. Among normal cells, apoptotic corpuscles could be detected by electron microscope. Quercetin inhibited the growth of human colon carcinoma cell line SW480 in a time-dependent and dose-dependent manner. Analysis of Flowcytometry results revealed that with the increase of quercetin using in SW480 cells, the apoptosis rate of human colon carcinoma cells was improved, and induced cell cycle arrest in G2/M. The adhesion experiments and Transwell method showed that quercetin significantly inhibit the adhesion and invasion of SW480 cells in does-dependent manner. Gelatin zymography study showed that quercetin treatment decreased the expressions of MMP-2 and MMP-9 in dose-dependent manner.Conclusions: In vitro, quercetin can efficiently inhibit the proliferation, adhesion, invasion and migration of SW480 cells, in a time-dependent and dose-dependent manner, and induce the apoptosis and cell cycle arrest of SW480 cells. Meanwhile, inhibit the expressions of MMPs may be one of the mechanisms underlying inhibition of invasion and migration by quercetin. So Quercetin is a potential drug to anti-invasion and anti-migration of tumor cells.
