Objective To investigate methylenetetrahydrofolate reductase (MTHFR) gene C677T polymorphism distribution of the Han and Hui people in Ningxia esophageal cancer( EC)and normal populations and to analyze MTHFR C677T polymorphism and its association with susceptibility to EC in Hui and Han people then compare with their difference, Simultaneously analyze the relationship between the environmental factors and esophageal cancer .Methods (1)Collect the whole blood specimens of 90 patients with EC in Ningxia,among the group 30 Hui cases and 60 Han cases while in the control group of 90 patients (Hui 30 cases and Han 60 cases). Hui people must be three generations to retain their national genetics, geography, customs, culture and life-style.(2)MTHFR C677T genotypes were detected by PCR- RFLP in 90 patients with EC (Hui 30 and Han 60)and 90 controls with no cancer in the same area(Hui 30 and Han 60)(.3)Theχ2 test was adopted to check the difference in genotype distribution between patients and controls, also check the increased risk for EC , check the accordance with Hardy-Weinberg equilibrium in the group.Results: (1) The frequencies of CC, CT, TT of MTHFR C677T in Han EC group in Ningxia is respectively 20%, 51.7%, 28.3%, and EC of Hui is 13.3%, 63.3%, 23.4% compared with no significant difference (χ2 = 1.178, P> 0.05);(2) In the normal control group, the frequencies of CC, CT, TT of MTHFR C677T of Han is 20%, 63.3%, 16.7% and Hui normal control group 46.7%, 50%, 3.3% were significantly different (χ2 = 8.436, P <0.05); (3) In Han EC group, MTHFR C677T CC, CT, TT respectively20%, 51.7%, 28.3%, with Han of the normal control group 20% , 63.3%, 16.7% was not significant difference (χ2 = 2.525, P> 0.05),but the EC TT genotype frequencies is higher than control group;(4) in Hui EC group, MTHFR C677T CC, CT, TT 13.3%, 63.3%, 23.4%, and Hui normal control group 46.7 %, 50%, 3.3% was not a significant difference (χ2=6.429,P<0.05);(5)The non-conditional logistic regression analysis showed that the increased risk for EC was significantly associated with a family history of gastrointestinal tumor(OR=3.613) , Bad eating habits ( OR=1.875 ), smoke or drink (OR=1.964);(6) The distribution of genotypes in the Han and Hui people are in line with Hardy-Weinberg genetic equilibrium equation (Hanχ2=3.338, P>0.05 Huiχ2=1.133, P>0.05). Conclusions (1) Distribution of MTHFR C677T polymorphism are different between Han and Hui normal population in Ningxia;(2) MTHFR C677T polymorphism in Hui people is related to the susceptibility of EC while nothing to do with Han people possibly;(3) Bad eating habits,smoking,drinking and a family history of gastrointestinal cancer are risk factors of EC, in which family history is most closely to EC.
